【摘要】:目的评估ISOLA全自动平板接种仪使用Digest-EUR与SPUTASOL、胰蛋白酶痰消化液处理痰标本的液化和接种效果,以及不同试剂对细菌生长的影响,为选择痰消化液提供依据。方法收集2014年10月-2015年1月医院住院患者120份中等粘稠痰标本和特粘稠痰标本,同时采用三种试剂对标本进行预消化处理,评估接种效果,三种试剂分别与肺炎链球菌、流感嗜血菌菌液接触0、15、30、60、90、120min,观察不同作用时间对肺炎链球菌和流感嗜血菌生长的影响,并使用SPSS统计软件进行统计分析。结果三种试剂对两种粘度痰标本消化处理后,进行全自动标本接种均未出现吸头堵塞,消化预处理效果差异无统计学意义;在哥伦比亚血平板、中国蓝平板和巧克力平板上的接种效果差异无统计学意义;Digest-EUR和SPUTASOL两种试剂与肺炎链球菌、流感嗜血菌菌液接触不同时间后,均对生长无明显影响,胰蛋白酶作用15min后,即对肺炎链球菌、流感嗜血菌的生长产生抑制。结论三种试剂均可用于ISOLA全自动平板接种仪对痰标本的消化预处理,其中Digest-EUR和SPUTASOL消化液对肺炎链球菌和流感嗜血菌生长无明显影响,胰蛋白酶液化15min后对肺炎链球菌和流感嗜血菌的生长有明显的抑制作用。
[Abstract]:Objective to evaluate the effect of liquefaction and inoculation of sputum samples treated with Digest-EUR and SPUTASOL with ISOLA automatic plate inoculation instrument. And the effects of different reagents on the growth of bacteria. Methods from October 2014 to January 2015, 120 cases of moderately viscous sputum and particularly viscous sputum were collected. At the same time, the samples were predigested with three reagents to evaluate the inoculation effect. The three reagents were exposed to Streptococcus pneumoniae and Haemophilus influenzae solution for 120min. The effects of different action time on the growth of Streptococcus pneumoniae and Haemophilus influenzae were observed and analyzed by SPSS software. No head clogging was found in all the fully automatic inoculated specimens, and there was no significant difference in the effect of digestion and pretreatment. There was no significant difference in inoculation effect between Chinese blue plate and chocolate plate in Colombian blood plate. After contact with Streptococcus pneumoniae and Haemophilus influenzae for different time, Digest-EUR and SPUTASOL had no obvious effect on growth. The growth of Streptococcus pneumoniae and Haemophilus influenzae was inhibited. Conclusion the three reagents can be used for digesting and pretreating sputum samples with ISOLA automatic plate inoculation instrument. Digest-EUR and SPUTASOL had no effect on the growth of Streptococcus pneumoniae and Haemophilus influenzae. Trypsin liquefaction for 15 min inhibited the growth of Streptococcus pneumoniae and Haemophilus influenzae.
