【摘要】:全基因组关联研究是指在全基因组层面上,利用大量的样本,对基因与疾病的关联性进行反复验证的一种研究.作为一种发现复杂疾病易感基因的重要工具,全基因组关联研究已经帮助许多研究人员成功的找到了数千个与多种疾病相关联的位点.相对于一步法而言,合理的两步法能够大幅度的降低测序工作量和费用.两步法使用同一个样本,并且在两步中分别使用两个不同的统计量.首先,我们对样本中大量的候选位点进行筛选,选出一些显著的位点.然后,对上一步筛选出来的那些位点进行进一步的检验,确定与目标性状相关联的基因位点.当前,两步法成为全基因组关联性研究中一种常见的方法.在家庭数据中,关联信息可以被划分为家庭之间的信息和家庭内部的信息.在第一步中,利用家庭之间的信息来筛选标记位点;在第二步中,利用家庭内部的信息对已选择的标记位点进行关联分析.本文对已有的两步法进行了深入的研究,并且提出了一种新的两步法.新方法基于核心家庭数据,利用两个不相关的统计量对基因位点进行筛选和检验,之后进行模拟研究,以判断新方法的第一类错误率和功效,并与其他方法进行比较.
[Abstract]:A genome-wide association study refers to the whole genome level, using a large number of samples, a study of repeated verification of relevance to gene and disease. As an important tool for complex disease susceptibility gene, genome-wide association studies have helped many researchers successfully found associated with many thousands of the disease locus. Compared with one step, two step reasonable can greatly reduce the workload and cost. The two step was to use the same sample, and respectively using two different statistics in two steps. First, we screened for candidate loci of large samples, select some significant sites then. Further, the inspection of those sites were screened out one step, determine the relevant and objective traits associated loci. At present, two step into genome-wide association studies A common method. In the family data, related information can be divided into family between information and information within the family. In the first step, to screening marker loci between families using information; in the second step, association analysis of marker loci have been selected by family information. In this paper, two step on the in-depth study, and puts forward a new two step method. The new method based on the core of family data, screening and testing of gene loci using two statistics are not related, after simulation study, the first class error rate and efficacy judgment method, and compared with other methods.
