利用可见近红外光谱分析技术的组织血氧饱和度无创检测方法

【摘要】:血氧饱和度指血液中氧合血红蛋白的含量占血红蛋白总量的百分比,人体组织血氧饱和度是指局部组织中各种微血管中血液的氧饱和度综合值,反映局部组织的氧合情况。无损、持续和快速监测人体组织中血氧饱和度具有十分重要的临床意义。本文在综述了血氧饱和度检测方法和光谱分析技术原理、发展现状的基础上,根据人体组织中血红蛋白的光学特性和光谱分析技术的特点,提出了一种利用可见近红外光谱分析技术的无创检测组织血氧饱和度的方法。主要研究工作内容如下:(1)根据氧合血红蛋白与脱氧血红蛋白在200~1000nm波段的吸收光谱特点,提出了一种利用可见近红外光谱技术检测血氧饱和度的方法,通过数据处理与建模手段对溶液中血红蛋白的血氧饱和度进行预测,从而可开发成无创检测组织中血液血氧饱和度的方法。(2)建立了血红蛋白氧合实验系统并优化了系统参数,利用血红蛋白氧合模型对上述可见近红外光谱方法预测的血氧饱和度进行了定标,并分析了其中的影响因素。通过建立的可见近红外光谱回归模型就能对预测集样本的血氧饱和度进行预测。(3)将所提出的方法应用于人体血液中的血氧饱和度检测。完成了血液红细胞氧合实验系统设计并优化了系统参数,在血液红细胞氧合体系中对上述可见近红外光谱方法预测的血氧饱和度进行了定标,并且对血液浓度等影响因素进行了分析。建立可见近红外光谱回归模型,能够对预测集样本的血氧饱和度进行预测,并对预测结果进行了分析。实验结果表明,在检测血红蛋白溶液中血氧饱和度时,该方法对血氧饱和度的预测相对误差能保持在6%以内;在检测人体血液中的血氧饱和度时,大部分样本点处预测相对误差在10%以内。综上所述,本文研究的可见近红外光谱无损检测组织血氧饱和度的方法能够有效地用透射方式检测溶液中的血氧饱和度,可以进一步开发设计成为人体组织血氧饱和度的无创检测装置。
[Abstract]:Blood oxygen saturation refers to the percentage of oxygenated hemoglobin in blood to the total hemoglobin, and oxygen saturation in human tissue is the comprehensive value of blood oxygen saturation in various microvessels in local tissues. Continuous and rapid monitoring of blood oxygen saturation in human tissues is of great clinical significance in reflecting the oxygenation of local tissues. This article reviews the measurement methods of blood oxygen saturation and the principle of spectral analysis. Based on the present situation, according to the optical properties of hemoglobin in human tissues and the characteristics of spectral analysis technology. A noninvasive method for detecting blood oxygen saturation of tissues using visible near infrared spectroscopy (VNIR) is proposed. The main contents of this study are as follows: 1). According to the absorption spectra of oxygenated hemoglobin and deoxyhemoglobin at 1000nm. A method of detecting oxygen saturation of blood by using visible near infrared spectroscopy is proposed. The oxygen saturation of hemoglobin in solution is predicted by means of data processing and modeling. Therefore, a non-invasive method for detecting blood oxygen saturation in tissues was developed.) the hemoglobin oxygenation experimental system was established and the system parameters were optimized. Hemoglobin oxygenation model was used to calibrate the oxygen saturation predicted by the visible near infrared spectroscopy. The influence factors were analyzed. By using the VNIR regression model, the oxygen saturation of the predicted sample can be predicted. The proposed method is applied to the measurement of blood oxygen saturation in human blood, and the system parameters of the blood red blood cell oxygenation experiment system are designed and optimized. The oxygen saturation predicted by the visible near infrared spectroscopy was calibrated in the blood erythrocyte oxygenation system, and the influence factors such as blood concentration were analyzed. The visible near infrared spectroscopy regression model was established. It can predict the oxygen saturation of the predicted sample and analyze the predicted results. The experimental results show that the oxygen saturation in hemoglobin solution is detected. The relative error of the method for predicting oxygen saturation can be kept within 6%. In the detection of oxygen saturation in human blood, the relative error of prediction at most of the sample points is less than 10%. In this paper, the method of non-destructive detection of tissue oxygen saturation by near infrared spectroscopy can effectively detect the oxygen saturation in solution by transmission method. It can be further developed and designed as a non-invasive measurement device for blood oxygen saturation in human tissues.
