肺炎克雷伯菌噬菌体LH-02的生物学特性及基因组初步研究

【摘要】:目的研究肺炎克雷伯菌噬菌体LH-02的生物学特性及基因组性质。方法观察LH-02的噬菌斑形态和电镜下形态;调查LH-02的噬菌谱,并观察其一步生长曲线以及理化稳定性;提取噬菌体基因组进行酶切鉴定。结果LH-02在宿主菌KF29菌苔上形成直径2~3mm的透明噬菌斑,外周有宽2~3mm的半透明晕环;电镜观察确定其为有尾噬菌体目,短尾噬菌体科;一步生长曲线显示LH-02感染KF29的潜伏期短(约15min),裂解量大(约217PFU/细胞);LH-02在一个较宽的pH范围内(5~10)和较高的温度(50℃)范围内活性稳定;LH-02的核酸可被KpnⅠ,EcoRⅠ,HindⅢ降解,但不能被BamHⅠ降解。结论 LH-02为裂解性噬菌体,基因组为双链DNA,其潜伏期短,裂解量大,稳定性好,有望成为生物抗菌剂。
[Abstract]:Objective to study the biological and genomic properties of Klebsiella pneumoniae phage LH-02. Methods the plaque morphology and electron microscope morphology of LH-02 were observed, the phage spectrum of LH-02 was investigated, the one-step growth curve and physicochemical stability were observed, the phage genome was extracted and identified by enzyme digestion. Results LH-02 formed transparent plaque with diameter 2~3mm on the coating of host strain KF29, and had a semitransparent halo ring with wide 2~3mm around it. The one-step growth curve showed that the incubation period of LH-02 infection with KF29 was short (about 15 mins), and the nucleic acid with stable activity of LH-02 could be degraded by Kpn 鈪,
