葡萄糖对铜绿假单胞菌生物膜形成能力的影响

【摘要】:目的探索不同浓度的葡萄糖对铜绿假单胞菌生物膜形成能力的影响。方法用含有不同葡萄糖浓度的胰蛋白胨大豆肉汤培养铜绿假单胞菌,测定其570 nm处光密度,通过函数Linest进行线性回归拟合,用激光扫描共聚焦显微镜(CLSM)观察不同浓度葡萄糖培养铜绿假单胞菌生物膜的形成过程及厚度。结果 22株细菌不同浓度葡萄糖的A_(570)拟合直线的斜率均大于0,其中4株菌上升趋势明显。与无糖组及低浓度葡萄糖组相比,当葡萄糖浓度≥11.1mmol·L~(-1)时,生物膜成熟速度加快、成熟时间减少、粘附能力增强、厚度增加(P0.05)。结论葡萄糖浓度≥11.1 mmol·L~(-1)时对铜绿假单胞菌生物膜形成有明显的诱导作用,并且随葡萄糖浓度的升高,诱导作用增强。

Methods Pseudomonas aeruginosa was cultured in soybean broth containing different glucose concentrations. The optical density at 570Nm was determined and linear regression fitting was carried out by functional Linest. The formation process and thickness of Pseudomonas aeruginosa biofilm cultured with different concentrations of glucose were observed by laser scanning confocal microscope (CLSM).
