免疫胶体金试纸检测仪的研究

【摘要】:免疫胶体金试纸检测已成为一种常规的检测方法,在生物医学、农牧业、环境监测、食品安全等方面都有非常广泛的应用。目前试纸检测结果的读取还处于依赖人眼目测的定性水平,无法实现准确的定量测量;此外,一些特殊检测需要结果实时留存,例如毒品检测等,不仅需要准确的结果,还需要提高检测效率。因此设计一款能够实现快速准确检测、可实时记录存储、兼容各类胶体金试纸的小型化便携分析仪成为市场需求。为解决试纸检测目前存在的部分问题,本论文设计了一款基于树莓派的免疫胶体金试纸检测仪。论文主要工作如下:(1)分析胶体金试纸的工作原理,结合国内外研究现状及设计理念,针对试纸的反应显色特性设计适宜的检测方案。(2)完成系统整体结构的搭建,介绍系统检测原理,并分别阐述了各个模块及其功能,主要包括:均匀照明光路、中央控制系统树莓派、摄像头、打印机和触摸屏。(3)介绍系统识别图像处理图像的具体方法。系统引入二维码对不同类型试纸进行区分,使用二维码识别不仅能实现试纸检测区定位,还能够扩大检测仪使用范围,使仪器实现多种试纸通用性。检测系统将采集到的试纸图像进行灰度化、滤波除噪、图像分割、二值化等操作,最终成功提取试纸显色带的灰度值数据。(4)通过调整LED工作电压与摄像头采集帧率进行实验,选择系统最佳实验条件。利用一组灰白标准色带对试纸的测量数据进行修正,使测量结果更接近于真实值。另外重复性测量表明系统具有较高的稳定性,变异系数(CV)控制在5‰以内。本论文设计的免疫胶体金试纸检测仪能不但够准确测量出试纸灰度值,而且可实时打印检测结果,操作简单方便,能够满足不同类型试纸的检测需求,具有良好的应用价值和发展前景。
[Abstract]:Immunocolloidal gold test paper has become a conventional detection method, which has been widely used in biomedicine, agriculture and animal husbandry, environmental monitoring, food safety and so on. At present, the reading of test paper test results is still in the qualitative level that depends on human eye measurement, so it is impossible to realize accurate quantitative measurement. In addition, some special tests need real-time retention of results, such as drug detection, not only need accurate results, but also need to improve detection efficiency. Therefore, it is a market demand to design a compact portable analyzer which can be used for fast and accurate detection, real-time recording and storage, and compatible with all kinds of colloidal gold test paper. In order to solve some problems existing in paper test, an immune colloidal gold paper tester based on raspberry pie was designed in this paper. The main work of this paper is as follows: (1) analyzing the working principle of colloidal gold test paper, combining the domestic and foreign research status and design idea, designing the suitable detection scheme for the reaction color characteristic of the test paper. (2) completing the construction of the whole system structure. This paper introduces the principle of system detection, and describes each module and its functions, including: uniform lighting, central control system, raspberry pie, camera, etc. Printer and touch screen. (3) the method of image processing is introduced. The system introduces the two-dimensional code to distinguish different types of test paper. Using the two-dimension code recognition can not only realize the location of test paper detection area, but also expand the scope of use of the tester, so that the instrument can realize the versatility of various test papers. The detection system performs grayscale, filtering and denoising, image segmentation, binarization and so on. Finally, the gray value data of the test paper color display band are extracted successfully. (4) by adjusting the working voltage of LED and the frame rate collected by the camera, the optimal experimental conditions of the system are selected. A group of gray and white ribbons are used to correct the measured data of the test paper, and the results are closer to the real values. In addition, the repeatability measurement showed that the system had high stability, and the coefficient of variation (CV) was controlled within 5 鈥,
