油菜籽蛋白质膜的制备与性能研究

【摘要】:菜籽蛋白是一类优质的植物蛋白资源,含有大量的分子间作用力,范德华力,二硫键以及疏水相互作用使得菜籽蛋白具有成膜的潜质。本文通过使用琼脂复配制备了性能优良的菜籽蛋白质薄膜,并对菜籽蛋白的提取过程以及最佳的成膜条件进行了探究,并对其在包装领域的应用上进行了初步的探究。以菜籽粕为原料,采用酸提碱沉淀的方法提取菜籽蛋白,并建立了利用菜籽蛋白结合色素自身显色的方法来测定菜籽蛋白的含量。以不同浓度的菜籽蛋白溶液绘制标准曲线,并用凯氏定氮法标定曲线,其1.0-3.0mg/m L范围内线性关系良好,线性相关系数R2大于0.999,平均回收率为99.1%,精密度,重现性良好,适用于大批量同品种菜籽蛋白的快速检测。以菜籽提取蛋白制备菜籽蛋白质薄膜,并对薄膜的性质进行了研究,结果显示,菜籽蛋白成膜溶液最佳的处理条件为甘油含量20%,菜籽蛋白含量为30%,处理温度50℃,p H10.0,紫外辐射处理24h。在此条件下,薄膜的抗拉强度为3.5MPa,断裂伸长率为20%。采用十二烷基硫酸钠改性的埃洛石纳米管对薄膜进行改性研究,结果发现,最佳的改性剂用量为3%,在此条件下薄膜的抗拉强度提高约20%,但是其断裂伸长率则降低约10%。微观结果测试显示,薄膜的层状结构已经改变为单层的薄膜,并且改性埃洛石纳米管通过物理吸附与化学交联的方式共同改善薄膜的性质。菜籽蛋白质与琼脂共混膜的最佳制备条件为菜籽蛋白与琼脂比为3:7,其他条件与制备菜籽蛋白膜相同,此时薄膜的抗拉强度和断裂伸长率分别为35MPa和10%,较单一的菜籽蛋白膜有大幅的提升;同时共混膜的透明度以及阻隔性能有了大幅的改善。结果说明,共混是改善蛋白质薄膜的性能的有效手段。薄膜的微观结构显示,共混后的菜籽蛋白膜具有更加致密的结构,这也说明了共混是提高蛋白质薄膜性能的有效手段。在对菜籽蛋白共混膜的改性研究中发现,使用对菜籽蛋白膜的安全性做了初步的探究,结果说明,菜籽蛋白有毒物质含量低于相关标准,薄膜在高温下不会挥发出有毒有害物质。
[Abstract]:Rapeseed protein (rapeseed protein) is a kind of high quality plant protein resource, which contains a lot of intermolecular force, van der Waals force, disulfide bond and hydrophobic interaction, which makes rapeseed protein have the potential of film formation. In this paper, rapeseed protein thin films with excellent properties were prepared by using Agar, the extraction process and the optimum conditions of film formation were studied, and the application of rapeseed protein in packaging field was preliminarily explored. Rapeseed protein was extracted from rapeseed meal by acid extraction and alkali precipitation. The method of coloring rapeseed protein binding pigment was established to determine the content of rapeseed protein. The standard curve was drawn with different concentrations of rapeseed protein solution and calibrated by Kjeldahl method. The linear correlation coefficient R 2 was greater than 0.999 9, the average recovery was 99.1%, the precision and reproducibility were good, the linear correlation coefficient R 2 was greater than 0.999 9, and the calibration curve was calibrated by Kjeldahl method. It is suitable for rapid detection of rapeseed protein of the same variety in large quantities. Rapeseed protein films were prepared by extracting protein from rapeseed, and the properties of the films were studied.
