温敏性纳米水凝胶颗粒的合成及对多肽的识别研究

【摘要】:温敏性纳米水凝胶是一类对温度敏感的高分子聚合物纳米水凝胶材料,可以溶于水或者水溶液,具有最低临界溶解温度。近年来,由N-异丙基丙烯酰胺(NIPAm)聚合而成的温敏性水凝胶引起人们的普遍关注。 本研究以NIPAm为结构框架和温敏性单体、丙烯酸(AAc)为亲电作用单体、N-叔丁基丙烯酰胺(tBAm)为疏水作用单体、N,N’-亚甲基双丙烯酰胺(BIS)为交联剂、十二烷基硫酸钠(SDS)为表面活性剂、N,N,N’,N’-四甲基乙二胺(Temed)为助联剂、过硫酸铵(APS)为引发剂、在水溶液中制备出一系列温敏性纳米水凝胶颗粒。经扫描电镜(SEM)和动态光散射仪(DLS)表征,,粒径均匀。实验考察了合成配方中SDS和AAc用量对颗粒粒径的影响。SDS用量从0~50mg变化,粒径变化为460~70nm。AAc从5%~40%(mol%),粒径变化为74~613nm,且电位绝对值增加,体系趋于稳定。纳米水凝胶颗粒在低于15℃时粒径较大并保持均一稳定状态,随着温度上升,尤其当温度上升至20-30℃之间时,颗粒粒径开始急速缩小。 将所制备的纳米水凝胶颗粒用于组氨酸标签多肽(NH2-LEVLFQGPHHHHHH-COOH/HOOC-LEVLFQGPHHHHHH-NH2)的识别,经高效液相色谱分析,纳米水凝胶颗粒粒径越小,对目标多肽吸附率越高;AAc (负电荷单体)摩尔浓度对吸附率影响较大,采用NIPAm/tBAm/AAc/BIS(43/40/15/2, mol%)配方所得纳米水凝胶颗粒对目标多肽吸附率最高,达78%;纳米水凝胶颗粒对目标多肽吸附率随着吸附溶液pH值下降而上升。 将所制备的纳米水凝胶颗粒用于蜂毒肽(NH2-GLY-ILE-GLY-ALA-VAL-LEU-LYS-VAL-LEU-THR-THR-GLY-LEU-PRO-ALA-LEU-ILE-SER-TRP-ILE-LYS-ARG-LYS-ARG-GLN-GLN-COOH)的识别,经高效液相色谱、聚丙烯酰胺凝胶电泳、紫外-可见分光光度计、细胞体外观察,随着纳米水凝胶粒径减小,其对蜂毒肽吸附作用逐渐升高;疏水单体tBAm的配比是影响吸附率的最重要因素,随着此单体配比的上升,其对蜂毒肽吸附率增大;AAc的配比对其吸附率影响不大。通过调节温度,利用纳米水凝胶的温敏性特征和其在不同粒径下对蜂毒肽吸附率的区别,可以控制纳米水凝胶颗粒对蜂毒肽吸附-脱附的过程。
[Abstract]:Temperature-sensitive nano-hydrogels are a kind of thermo-sensitive polymer nano-hydrogels which can dissolve in water or aqueous solution and have the lowest critical dissolution temperature. In recent years, thermosensitive hydrogels polymerized by N-isopropyl acrylamide (NIPAm) have attracted much attention. In this study, NIPAm was used as the structure frame and temperature sensitive monomer, acrylic acid (AAc) as electrophilic monomer, N- tert-Ding Ji acrylamide (tBAm) as hydrophobic monomer, and NNT-methylene bisacrylamide (BIS) as crosslinking agent. A series of thermo-sensitive nanogel particles were prepared in aqueous solution with sodium dodecyl sulfate (SDS) as surfactant, NNX (N (N) (n () (n) (n () The particle size was uniform by scanning electron microscope (SEM) and dynamic light scattering (DLS). The effects of the amount of SDS and AAc on the particle size were investigated. The content of SDS changed from 0~50mg to 540% (mol%) for 460~70nm.AAc, and the particle size changed from 74 ~ 613 nm. The absolute value of potential increased and the system tended to be stable.
