【摘要】:荧光分析法由于具有灵敏度高、信噪比高、成本低、操作简单等优点而被人们广泛关注。大部分的荧光探针分子可以用于细胞成像,有些甚至可以实现生物小分子的识别、生物体内特定蛋白质的检测以及DNA的测序等,这些技术为生物、医学、环境等领域的研究提供了很大的帮助。因此,寻找一种选择性好、灵敏度高、细胞渗透性和相容性好、细胞毒性小、对目标分子能够特异性识别的荧光染料分子具有非常重要的意义。氟化硼络合吡咯二甲川(简称BODIPY)类荧光探针,其具有结构可修饰性好,光稳定性好,有较高的荧光量子产率和摩尔消光系数,半峰宽窄等优点而广泛地被人关注。然而传统的BODIPY分子以吡咯为起始原料,合成产率低,分离困难,限制了其发展。本文在文献的基础上,合成了一类新型的类BODIPY荧光分子,完成了以下的工作:1.设计合成了一系列高选择性的反应型识别汞离子的荧光探针分子(C1-C4,图1)。并利用汞离子的嗜硫性,基于下列反应(图2),C1-C4可以在水溶液中实现对汞离子的特异性识别,响应速度快,选择性好。细胞实验表明,该类染料分子具有很好的细胞渗透性,低毒,可以用于活体细胞中Hg2+的检测。2.制备了氨基修饰的类BODIPY的荧光分子L(式1),利用桑德迈尔反应,成功实现了酸性条件下识别NO2-分子,识别特异性好,识别速度快,具有很好的应用前景。
[Abstract]:Because of its high sensitivity, high signal-to-noise ratio, low cost and simple operation, fluorescence analysis has been widely concerned. Most fluorescent probe molecules can be used in cell imaging. Some can even realize the recognition of biological small molecules, the detection of specific proteins in organisms and the sequencing of DNA. These technologies have provided great help for the research of biology, medicine, environment and so on. Look for a kind of good selectivity, high sensitivity, good cell permeability and compatibility, little cytotoxicity. It is very important for the fluorescent dye molecules which can be specifically recognized by the target molecule. Boron fluoride complex pyrrole dimethylChuan (BODIPYY) fluorescent probe has good structural modifiability. Light stability, high fluorescence quantum yield and molar extinction coefficient, narrow half-peak width and other advantages have been widely concerned. However, the traditional BODIPY molecules with pyrrole as the starting material, the synthesis yield is low. In this paper, a new class of BODIPY like fluorescent molecules has been synthesized based on the literature. A series of highly selective fluorescent probe molecules, C1-C4, were designed and synthesized. Based on the following reactions (Fig. 2) C1-C4 can achieve specific recognition of mercury ions in aqueous solution, with fast response and good selectivity. This kind of dye molecules have good cell permeability and low toxicity, and can be used to detect Hg2 in living cells. 2. The fluorescent molecule L (Formula 1) of amino-modified BODIPY was prepared. The Sundmayer reaction was used to realize the recognition of no _ 2-molecule in acidic condition. It has good recognition specificity and fast recognition speed. It has a good prospect of application.
