取代型多金属氧酸盐在显色免疫检测中的性能研究

【摘要】:显色免疫检测是通过体系颜色的变化有针对性的检测某种物质,在分析中通常会用到生物酶,比如辣根过氧化物酶(HRP),但其容易受到外界环境的影响而变质。多金属氧酸盐(Polyoxometalates,POMs)是一类具有氧化还原性的金属-氧簇化合物,具有类似纳米的结构,其催化氧化性能可以应用于很多方面,包括显色免疫检测等。本论文基于POMs可以活化H_2O_2的性质,拟设计H_2O_2的HRP模拟化合物,并代替HRP。前列腺癌严重困扰着男性患者,在初期检测中经常用到的方法有:直肠指检、直肠超声、前列腺穿刺活检、电子计算机断层扫描(CT)和多重内反射(MIR)等,这些检测手段不仅会对患者造成极大的心理负担以及生理伤害,还会导致对病情的误判或过度治疗,所以发展一种高效便捷、具有人文关怀的方法迫在眉睫。无良商家在奶粉中添加三聚氰胺的事件,让社会对孩子食用的奶制品的质量存在质疑。因此,我们希望设计一种方便快捷、简单有效的方法来检测三聚氰胺,使食品检测更加切实可行,使食品安全得到保障。因此,我们合成一系列不同金属取代的杂多酸盐,研究其通过活化H_2O_2氧化有机底物TMB(C_(16)H_(20)N_2)的性能,探究其催化活性与氧化性之间的关系,并由此研究通过其性能检测肌氨酸和三聚氰胺的方法。1、制备不同金属取代的硅钨杂多酸盐:[SiW_(11)O_(39)M(H_2O)]~(6-)、[SiW_(10)O_(36)M_2(H_2O)_2]~(4-)、[SiW_9O_(37)M)3(H_2O)_3]~(8-),(M=Cu~(2+),Co~(2+),Mn~(2+),Fe~(2+));稀土金属取代的[SiW_(11)O_(37)L(H_2O)]n-(L=La~(3+),Ce~(4+));并利用红外光谱(IR)、紫外光谱(UV)、循环伏安(CV)等方法对其结构进行表征,并比较其氧化还原性能;研究其催化检测肌氨酸的活性,探究其线性关系,建立基于SiW_9Mn3快速检测肌氨酸的方法,检测线性范围为2.40×10-2~7.86×10~(-4) mol/L,最低检测限为4.96×10-3mol/L。2、制备不同金属取代的磷钨杂多酸盐:[PW_(11)O_(39)M]~(5-),[PW_9O_(34)M_3]~(3-)(M=Cu~(2+),Co~(2+),Mn~(2+),Fe~(2+));并利用IR、UV、CV等方法对其结构进行表征,并比较其氧化还原性能;研究其催化检测三聚氰胺活性,探究其线性关系,建立基于PW_(11)Co快速检测三聚氰胺的方法,检测线性范围为1.0×10~(-4)-1.5×10-6mol/L,最低检测限为2.5×10-6mol/L。
[Abstract]:Colour immunoassay is the specific detection of a substance by changing the color of the system, and bioenzymes are commonly used in the analysis. Horseradish peroxidase (HRP), for example, is susceptible to deterioration due to the external environment. Polyoxometales polyoxometalates (POMs) are a kind of redox metal-oxygen cluster compounds with similar nanostructures. The catalytic oxidation performance can be used in many fields, including color immunoassay. Based on the property that POMs can activate Hs _ 2O _ 2, this paper intends to design a HRP mimic compound of H _ 2O _ 2 and replace HRP.Prostate cancer seriously affects male patients. The methods often used in early detection include rectal digital examination, rectal ultrasound, prostate biopsy, computed tomography (CT) and multiple internal reflex (mir), etc. These testing methods will not only cause great psychological burden and physiological harm to patients, but also lead to misjudgment or overtreatment of the condition, so develop an efficient and convenient method. There is an urgent need for humane care. The incident of adding melamine to milk powder by unscrupulous merchants has raised questions about the quality of dairy products consumed by children. Therefore, we hope to design a convenient and fast one. A simple and effective method to detect melamine, make food detection more feasible and ensure food safety. Therefore, we synthesize a series of heteropolyacids substituted by different metals. To study its performance by activating the organic substrates of H _ 2O _ 2 oxidation, TMB-C _ s _ s _ 16 / T _ T _ I _ _ _ 2) and to explore the relationship between its catalytic activity and its oxidizing properties, And through the method of detecting creatine and melamine, we have studied the preparation of silicotungsten heteropoly salt substituted by different metals: [SiWA11ON39Mh _ 2O)] 6-C, [SiW_(10)O_(36)M_2(H_2O)_2] 6-C, [SiW_9O_(37)M)3(H_2O)_3] 8-MCuCu2MnCoC _ (2) M _ (2) MCuC _ (2) MN _ (2) O _ (2) H _ (2) O _ (2); rare earth metal-substituted [Siw _ (11) O _ (11) O _ (11) O _ (H _ 2O))] n-L Lahuo _ (3) (n ~ (1)) -L ~ (+) ~ (1) (n) -L ~ ((1)) -lLaA-1 / C ~ (3) C ~ (+ +) ~ (-) ~ (3) ~ (-) ~ (-) ~ (-) ~ (?). The structure was characterized by ACV and other methods. Its redox performance was compared, the activity of its catalytic detection of sarcosine was studied, the linear relationship was explored, and a method for rapid detection of sarcosine based on SiW_9Mn3 was established. The linear range of detection is 2.40 脳 10 ~ (-2) ~ 7.86 脳 10 ~ (-4) mol / L, and the lowest detection limit is 4.96 脳 10 ~ (-3) mol 路L ~ (-2). Different metal-substituted phosphotungstophosphorus heteropolyates: [PW_(11)O_(39)M] -5 ~ (-), [PW_9O_(34)M_3] ~ (3) ~ (-) ~ ((1)) ~ (2) CoCo22 ~ (2) mn ~ (2) mn ~ (2) C ~ (2) C ~ (2) C ~ (2) C ~ (2) is used to characterize its structure, and to compare its redox properties, and to study its catalytic activity for the detection of melamine, and to study its catalytic activity. The linear relationship was investigated and a rapid detection method for melamine based on PW_(11)Co was established. The linear range was 1.0 脳 10 ~ (-4) -1.5 脳 10 ~ (-6) mol / L, and the minimum detection limit was 2.5 脳 10 ~ (-6) mol / L.
