基于化学衍生化的靶向脂质组学质谱分析研究进展

【摘要】:脂质组学研究与一些重大疾病(如心血管病、糖尿病、肿瘤等)密切相关,是当今生命科学研究的重要领域.质谱分析技术是目前脂质组学研究的核心手段之一.化学衍生化策略可显著提高基于鸟枪法或液相色谱-质谱联用方法的脂质组学分析性能,近年来已有一些相关研究报道.本文重点介绍了基于化学衍生化的质谱分析方法在靶向脂质组学研究中的进展,并提出了问题和展望.希望在衍生化试剂的设计与合成,特别是具有显著优势的稳定同位素标记衍生试剂开发,以及新的衍生化策略和相应的质谱分析方法的开发与应用方面起到一定的推动作用.
[Abstract]:Lipid studies are closely associated with major diseases such as cardiovascular disease, diabetes, cancer, etc. Mass spectrometry (MS) is one of the core methods of liposome research. The chemical derivatization strategy can significantly improve the performance of liposome analysis based on shotgun method or liquid chromatography-mass spectrometry (LC-MS). In recent years, some related studies have been reported. This paper focuses on the development of mass spectrometry based on chemical derivation in the study of targeted liposome, and puts forward some problems and prospects. It is hoped that the design and synthesis of derivatization reagents will be carried out. In particular, the development of stable isotope labeled derivatives, as well as the development and application of new derivatization strategies and the corresponding mass spectrometric analysis methods, have played a certain role in promoting the development and application of stable isotope labeled derivatives.
