UPLC法同时测定连参通淋片中5种黄连生物碱类成分的含量

【摘要】：目的:建立同时测定连参通淋片中表小檗碱、盐酸药根碱、黄连碱、盐酸巴马汀和盐酸小檗碱含量的方法。方法:采用超高效液相色谱法。色谱柱为Thermo Scientific Syncronis C_(18),流动相为乙腈-0.1%磷酸溶液(加三乙胺调p H至3.0)(30∶70,V/V),流速为0.3 mL/min,检测波长为345 nm,柱温为25℃,进样量为2μL。结果:表小檗碱、盐酸药根碱、黄连碱、盐酸巴马汀和盐酸小檗碱检测进样量线性范围分别为4.08~163 ng(r=0.999 9)、5.72~229 ng(r=0.999 8)、5.67~227 ng(r=0.999 9)、6.78~271 ng(r=0.999 6)、13.2~526 ng(r=0.999 9);精密度、稳定性、重复性试验的RSD2.0%;加样回收率分别为96.83%~103.92%(RSD=2.2%,n=9)、97.62%~104.85%(RSD=2.9%,n=9)、99.44%~104.18%(RSD=1.8%,n=9)、98.64%~104.42%(RSD=1.8%,n=9)、98.24%~102.40%(RSD=1.5%,n=9)。结论:该方法简便、灵敏、快速、准确,适用于连参通淋片中5种黄连生物碱类成分含量的同时测定。
