液质联用法测定不同产地五味子中七种有效化学成分的含量

【摘要】:目的建立五味子中五味子醇甲、五味子醇乙、五味子酯甲、五味子酚、五味子甲素、五味子乙素、五味子丙素的含量测定方法。方法采用液相色谱质谱联用法,以色谱柱ACQUITY UPLC BEH C_(18)(2.1 mm×50 mm,1.7μm),流动相为乙腈(A)-0.1%甲酸水溶液(B),梯度洗脱0~5 min,A为40%;5~7 min,A为40%~70%;7~10 min,A为70%;10~11 min,A为100%;11~12 min A为40%,回到初始条件,流速为0.3 ml/min,进样量2μl,柱温30℃,ESI离子源在正离子模式下选择离子扫描模式对五味子中的七种木脂素进行定量分析。结果七种木脂素在一定范围内线性良好,相关系数均大于0.999,加样回收率在94.48%~96.70%之间,RSD在0.58~4.07之间,n=9。结论该方法快速、灵敏度高、选择性好,适合测定五味子中木脂素的定量分析。
