长药八宝的抗氧化活性及其有效成分的分离研究

【摘要】:衰老是当今威胁人类生命健康最主要的原因之一。氧化性自由基的积累加速了机体的衰老。自由基引起的氧化应激与众多疾病的发生、发展有重大关系[1],使得寻找"完美"抗氧化剂的任务日益艰巨。在营养物质普遍回归大自然的热潮中,天然物宝库既具有丰富的营养物质,又作为抗氧化剂最具潜力的来源之一,受到了人们的广泛关注。天然抗氧化剂具有安全、可靠、生理活性多等优点被人们普遍接受,引起人们浓厚的研究兴趣。八宝属植物具备缓解疲劳、清热、解毒、活血化瘀、消肿止痛之效。近年来,人们对八宝属植物的药用价值的关注与日俱增,以期充分开发八宝属植物的利用价值。长药八宝(Hylotelephiumspectabile)为景天科八宝属,多年生肉质植物草本。地以上的茎粗壮、直立,根肉质、短粗,具有显著的耐旱性。可治疗跌打损伤,蚊虫叮咬,疱疹,脚癣等。目前,人们对长药八宝的研究主要集中于作为观赏植物,对其生物活性研究甚少。本文以长白山地区长药八宝的地下根部分为研究对象,采用浸泡和回流两种方式对其活性物质进行提取。提取物经过一系列溶剂依次萃取,得石油醚层组分、三氯甲烷层组分、乙酸乙酯层组分、正丁醇层组分、水层组分。以Vc做阳性对比,测定各萃取相消除1,1-二苯基-2-苦肼基(DPPH)的能力,来研究长药八宝的抗氧化活性。结果表明:回流粗取物和浸泡粗提物的抗氧化活性均以乙酸乙酯层组分三氯甲烷层组分石油醚层组分正丁醇层组分水层组分顺序递减。但回流粗提物的抗氧化活性高于浸泡粗提物的抗氧化活性。尤其是回流提取物中乙酸乙酯组分,即便浓度在0.1mg/mL下,清除率也能到达96%,其IC50为0.05mg/mL,其抗氧化能力与维生素C(Vc)的相当。通过薄层析、柱层析对长药八宝的乙酸乙酯层的化学成分进行了分离研究。用核磁共振技术、液质联用、红外光谱方法鉴定了分离出的化合物的结构,首次从长药八宝中得到1,2-丙二醇、反式对羟基肉桂酸、顺式对羟基肉桂酸、异香草酸、2-甲基吡咯烷酮、对羟基苯甲酸、没食子酸。用紫外光谱法,对反式对羟基肉桂酸、对羟基苯甲酸、没食子酸的含量进行了测定。综上所述,长药八宝具有良好抗氧化活性,将为其应用于食品工业、化妆品行业及医疗保健品等领域,提供数据支持。
[Abstract]:Aging is one of the main reasons that threaten human life and health. The accumulation of oxidizing free radicals accelerates the aging of the body. The oxidative stress caused by free radicals is closely related to the occurrence and development of many diseases, which makes the task of searching for "perfect" antioxidants increasingly arduous. In the upsurge of nutrient returning to nature, natural treasure house is not only rich in nutrients, but also one of the most potential sources of antioxidants. Natural antioxidants are widely accepted by people because of their safety, reliability and many physiological activities. Bubao plants have the effect of relieving fatigue, clearing heat, detoxifying, promoting blood circulation and removing blood stasis, detumescence and pain. In recent years, people pay more and more attention to the medicinal value of the genus Euphorbia, in order to fully exploit the utilization value of the genus Euphorbia. Hylotelephium spectabile is a perennial fleshy plant herb. Stems above ground are stout, erect, rooting fleshy, short and tolerant to drought. Can be used to treat injuries, mosquito bites, herpes, tinea pedis and so on. At present, the research on the long medicine Bubao is mainly focused on the ornamental plant, but its biological activity is very little. In this paper, the underground roots of Changbaobao in Changbai Mountain area were studied. The active substances were extracted by soaking and reflux. The extract was extracted by a series of solvents in turn to obtain the components of petroleum ether layer, trichloromethane layer, ethyl acetate layer, n-butanol layer and water layer. The antioxidant activity of Babao was studied by using VC as positive contrast to determine the ability of each extraction phase to eliminate 1 ~ (-1) diphenyl-2-picrylhydrazinyl DPPHs. The results showed that the antioxidant activity of crude refluxing extract and soaking crude extract was decreased in the order of ethyl acetate layer, trichloromethane layer, petroleum ether layer, n-butanol layer, water layer component. However, the antioxidant activity of reflux crude extract was higher than that of soaking crude extract. In particular, the ethyl acetate fraction in the reflux extract, even if the concentration was at 0.1mg/mL, the clearance rate could reach 96mg / mL, its IC50 was 0.05mg / mL, and its antioxidant capacity was equal to that of vitamin C / Vc. The chemical constituents of ethyl acetate layer of Bubao were separated by thin chromatography and column chromatography. The structures of the compounds were identified by NMR, GC-MS and FTIR. For the first time, 1m2-propanediol, trans-p-hydroxycinnamic acid, cis-p-hydroxycinnamic acid were obtained from Babao. Isovanilic acid 2-methyl pyrrolidone, p-hydroxybenzoic acid, Gallic acid. The contents of trans-p-hydroxycinnamic acid, p-hydroxybenzoic acid and Gallic acid were determined by UV spectrophotometry. To sum up, Bubao has good antioxidant activity, which will provide data support for its application in food industry, cosmetics industry and medical and health products.
