用于检测次氯酸根和汞离子的荧光探针的设计合成及应用

【摘要】:荧光分析法操作简单,灵敏度高,能实现生物体内的实时动态监测,在医学、生命科学以及环境科学等领域得到广泛的应用。本论文正是基于荧光检测法设计合成了三种分别检测HClO和Hg~(2+)的荧光探针:1.我们设计合成一种基于2-(2'-羟基苯基)苯并噻唑(HBT)的新型“turn-on”型Cl O-荧光探针PHC2。基于ClO~-选择性氧化断裂偶氮键,探针对ClO~-显示出高度特异性选择性和高灵敏度。2.我们合成了叠氮基取代的2-(2'-羟基苯基)苯并噻唑(HBT)的“turn-on”型ClO~-荧光探针PHC3。探针在PBS:EtOH(V/V=8:2,pH 7.4,1%MeCN)的体系下对ClO~-有着良好特异性和灵敏度。3.我们以香豆素为荧光团,硫缩醛为报告基团而设计合成了“比率计”型荧光响应的汞离子探针PHC4。在PBS(pH 7.4)的检测体系下,体系呈现蓝色荧光,而随着Hg~(2+)的加入,体系在497 nm处的蓝色荧光峰逐渐降低,而在566 nm处产生新的黄色荧光峰,并不断增强,形成明显的比率计型荧光响应。此外,探针还实现了在海拉细胞中汞离子的荧光成像。
[Abstract]:Fluorescence analysis is simple in operation and high in sensitivity. It can be used in real-time dynamic monitoring of organisms and has been widely used in medicine, life science and environmental science. In this paper, we have designed and synthesized three fluorescent probes for detecting HClO and Hgnion 2, respectively, based on the fluorescence detection method. We have designed and synthesized a novel "turn-on" type ClO- fluorescence probe PHC _ 2 based on 2-butadiene-hydroxyphenyl) benzothiazolium (HBT). Based on the cleavage of azo bond by selective oxidation, the probe showed high selectivity and sensitivity to ClO-. We have synthesized the "turn-on" type ClOI-fluorescence probe PHC3 of azido-substituted 2-oxo-hydroxyphenyl) benzothiazolium (HBT). The probe has good specificity and sensitivity to ClO- in the system of PBS: EtOHN / V / V = 8: 2, pH 7.4 ~ (-1) MeCN-1). Using coumarin as the fluorescence group and thioacetal as the reporting group, we have designed and synthesized a "ratio" type mercury ion probe PHC4 with fluorescence response. The blue fluorescence was observed in the system of PBSX pH 7.4). The blue fluorescence peak at 497 nm decreased and the yellow fluorescence peak was increased at 566nm with the addition of Hgf2). An obvious ratio of fluorescence response is formed. In addition, the probe also realized the fluorescence imaging of mercury ions in Haila cells.
