含单个色氨酸的多肽分子的荧光特性研究:pH及金属离子的响应
[Abstract]:Because of the importance of metal ions, including copper, to the human body, A novel polypeptide molecule (WDAHSS) containing a single tryptophan is reported. It is proved that the novel molecule can be used to detect the fluorescence of copper ion by fluorescence spectrum. The intrinsic fluorescence contribution of the tryptophan residue contained by the molecule can be determined by copper ion sensitive detection. It is easy to be quenched by copper ion. The fluorescence spectra of copper ions interacting with WDAHSS at different pH values were analyzed and compared with those of a single tryptophan molecule. The mechanism of copper ion quenching of WDAHSS fluorescence was studied in detail. The results showed that the histidine in WDAHSS structure acted with copper ion through metal coordination and formed a solid square plane structure combined with peptide bond, which resulted in fluorescence quenching of tryptophan residues. The effects of different pH values on the fluorescence spectra of WDAHSS were discussed in detail. The binding constants of WDAHSS and copper ions were estimated by fluorescence spectrum measurement and numerical fitting. In order to enhance the ability of WDAHSS to resist pH interference, the amino terminal acetylation of WDAHSS was specially made, and the fluorescence emission was stabilized in the range of physiological pH. In addition, WDAHSS also adopts some specially designed structures to enhance its specific selectivity and biocompatibility for sensitive detection of copper ions. Further research on WDAHSS is expected to be used in vivo or in vivo fluorescence imaging.
