基于化学指纹图谱和多指标测定的柿叶药材质量研究

【摘要】:目的建立柿Diospyros kaki叶的HPLC指纹图谱,并进行多成分测定,为柿叶及其相关产品的质量控制提供科学依据。方法采用HPLC法,Diamonsil C18(250 mm×4.6 mm,5μm)色谱柱;以乙腈-0.2%甲酸水为流动相梯度洗脱;体积流量1.0 m L/min;柱温35℃;检测波长360 nm;进样量10μL;对22批柿叶药材进行指纹图谱研究,用中药色谱指纹图谱相似度评价系统(2004A版)进行评价,系统聚类分析和PCA分析将22批柿叶药材分为两类,同时对金丝桃苷、异槲皮苷、槲皮素和山柰素成分测定方法进行方法学考察。结果建立了柿叶的指纹图谱,共标定了12个色谱峰为柿叶药材指纹图谱的特征峰,金丝桃苷在0.044~0.880μg,异槲皮苷在0.078~1.560μg,槲皮素在0.040~0.800μg,山柰素在0.035~0.700μg线性关系良好,r均大于0.999 8,平均回收率分别为97.59%、98.97%、100.55%、99.96%。结论 HPLC指纹图谱结合多成分测定可为柿叶及其相关产品的质量控制提供参考。
