延胡索HPLC指纹图谱研究及9种生物碱含量测定

【摘要】:目的:分别建立延胡索药材及醋延胡索饮片的HPLC指纹图谱,同时对其中9种生物碱成分进行含量测定。方法:以通过三乙胺调节pH值至6.1的乙腈-0.1%磷酸溶液作为流动相,选择ECOSIL 120-5-C18AQ色谱柱(250 mm×4.6 mm,5.0μm)进行梯度洗脱。结果:延胡索药材和醋延胡索饮片各10批分别所得的指纹图谱,相似度值均0.94,确定共有峰均有15个。原阿片碱、盐酸巴马汀、盐酸小檗碱、脱氢紫堇碱、海罂粟碱、延胡索乙素、四氢小檗碱、延胡索甲素、脱氢海罂粟碱9种生物碱在相应范围内与其峰面积均呈良好线性关系(r≥0.9999);其平均加样回收率(n=6)分别为99.0%、101.5%、99.0%、102.2%、98.7%、100.3%、101.8%、99.4%、99.8%,RSD≤2.5%。结论:该方法具有较高的灵敏度,良好的重复性,可为延胡索药材和醋延胡索饮片的质量评价和全面控制提供参考依据。
