脂质体平衡透析与液质联用技术筛选脊痛宁片物质基础研究

【摘要】:目的研究脊痛宁片有效成分与模拟生物膜的相互作用,预测药物在体内的吸收情况。方法将脂质体作为模拟生物膜,采用脂质体平衡透析与液质联用技术筛选脊痛宁片中与脂质体有相互作用的成分,并考察脂质体质量浓度和缓冲液p H值对脊痛宁片与脂质体互相作用的影响。结果脊痛宁片中共有11个成分与脂质体作用明显,包括4个黄酮类成分,2个萜类成分,4个香豆素类成分,1个苯丙酸类成分。脂质体的质量浓度和缓冲液p H值都对脊痛宁片与脂质体互相作用有一定的影响。结论脂质体平衡透析与液质联用可预测脊痛宁片潜在的活性成分在体内的吸收情况,为其药效物质基础的进一步研究提供依据。
[Abstract]:Objective to study the interaction between the active components of Jitongning tablets and mimic biofilm and to predict the drug absorption in vivo. Methods Liposome was used as a mimic biofilm and liposome equilibrium dialysis and liquid-mass spectrometry were used to screen the components of Jitongning tablet that interact with liposome. The effects of liposome concentration and buffer pH on the interaction between Jitongning tablets and liposomes were investigated. Results there were 11 components in Jitongning tablets, including 4 flavonoids, 2 terpenoids, 4 coumarins and 1 phenylalanic acid. The mass concentration of liposome and pH value of buffer had some influence on the interaction between Jitongning tablets and liposomes. Conclusion Liposome equilibrium dialysis combined with liquid-mass spectrometry can predict the absorption of potential active components in Jiitongning tablets in vivo, which provides a basis for further study on the basis of its pharmacodynamic substance.
