UPLC-Q-TOF法分析双丹明目胶囊化学成分

【摘要】:目的通过超高效液相色谱-四级杆-飞行时间质谱(UPLC-Q-TOF-MS)法分析双丹明目胶囊化学成分。方法该胶囊内容物的分析采用Agilent Extend C18色谱柱(2.1 mm×50 mm,1.8μm);流动相0.1%甲酸(A)-乙腈(B),梯度洗脱;体积流量0.4 m L/min。根据对照品的相对分子质量、质谱碎片离子信息,结合参考文献对成分进行鉴定。结果从中共鉴定出102种成分,包括酚酸、黄酮及其苷类、萜类、香豆素、木脂素等。其中,24种来自旱墨莲,13种来自女贞子,11种来自丹参,14种来自泽泻,9种来自山茱萸,9种来自茯苓,6种来自牛膝,6种来自三七,4种来自牡丹皮,3种来自红土茯苓,3种来自山药。结论该方法灵敏、可靠、快速,可为双丹明目胶囊药效物质基础及质量控制研究提供依据。
[Abstract]:Objective to analyze the chemical constituents of Shuangdan Mingmu capsule by ultra-high performance liquid chromatography-4-step-time-of-flight mass spectrometry (UPLC-Q-TOF-MS). Methods Agilent Extend C18 column (2.1 mm 脳 50 mm,1.8 渭 m);) was used to analyze the contents of the capsule. The mobile phase of 0.1% formic acid (A) acetonitrile (B), was gradient eluted, and the volume flow rate was 0.4 mL / min. The components were identified according to the relative molecular weight of the reference substance and the information of fragment ion in mass spectrometry. Results 102 compounds were identified, including phenolic acid, flavonoids and their glycosides, terpenes, coumarins, lignans, etc. Of these, 24 were from Artemisia, 13 from Ligustrum lucidum, 11 from Salvia miltiorrhiza, 14 from alisma, 9 from Fructus Corni, 9 from Poria cocos, 6 from Achyranthes bidentata, 6 from Panax Notoginseng, and 4 from peony bark. Three species came from Rhizoma Poria cocos and three from Chinese yam. Conclusion the method is sensitive, reliable and rapid, and can provide basis for the study of the pharmacodynamics and quality control of Shuangdan Mingmu capsule.
