重楼HPLC指纹图谱及4种重楼皂苷含量测定

【摘要】:目的建立重楼药材的HPLC指纹图谱,并测定4种重楼皂苷的含量。方法采用HPLC法,Thermo BDS Hypersil C18(4.6mm×250mm,5μm)色谱柱,以乙腈-水为流动相梯度洗脱,流速为0.8 ml·min-1,检测波长为203 nm,柱温为30℃。对10批重楼药材进行分析,建立重楼药材的指纹图谱,同时测定了重楼皂苷Ⅰ ,Ⅱ , Ⅵ ,Ⅶ 4个成分的含量。结果初步建立了重楼HPLC特征指纹图谱共有模式,标定了13个共有峰。通过对照品归属法,确定了重楼皂苷I、重楼皂苷II、重楼皂苷VI和重楼皂苷VII共4个化合物,10批重楼相似度在0.851~0.970之间。4种重楼皂苷的含量测定线性良好(r0.9995,n=5),重楼皂苷I,II,VI,VII的平均回收率为99.41%,99.51%,94.77%,96.57%,RSD分别为1.20%,1.27%,2.55%,1.32%。结论该方法简便快捷,重复性好,可以用于重楼药材的鉴定与评价,为控制重楼药材的质量提供可靠的科学依据。
