续断HPLC特征图谱及3种活性成分的含量测定

【摘要】:目的:建立同时测定续断中川续断皂苷Ⅵ、马钱苷、当药苷3种活性成分含量的方法,并建立续断药材HPLC特征图谱。方法:采用Thermo BDS Hypersil C_(18)(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%磷酸溶液为流动相进行梯度洗脱,流速1.0 mL/min,柱温30℃,检测波长220 nm。结果:不同产地10批续断样品的HPLC谱图具有很好的一致性,标定了12个共有峰;地理位置相邻的鄂西南、湘西北共有8批样品的HPLC图谱相似度大于0.94。各样品的HPLC指纹图谱相似度结果与3种活性成分总量聚类分析结果一致。结论:所建立的HPLC特征图谱和含量测定方法能较好地评价和控制续断药材的质量,从指纹图谱化学成分信息的角度验证了包括五峰在内的鄂西南(包括临近的湘西北地区)所产续断的道地性或优质性。
