分离自安徽铜陵酸性矿山环境的AMDY2-9-2和AMDSY3-6-1还原铁的研究

【摘要】:本论文对分离自安徽铜陵酸性矿山环境中的两株铁还原菌进行了生物学鉴定和还原铁研究。通过对安徽铜陵酸性矿山环境中的野外样品(酸矿水、尾砂、排水沟底泥)进行采集,用选择性培养基FeT培养样品,经过分离纯化获得两株铁还原微生物AMDY2-9-2和AMDSY3-6-1。对两株微生物的16SrDNA测序和同源性分析表明,菌株AMDY2-9-2与A cidithiobacillus thiooxidans亲缘关系最为接近,同源性为99.79%,而AMDSY3-6-1与Acidithiobacillus ferridurans亲缘关系最为接近,同源性为95.78%。菌株AMDY2-9-2和AMDSY3-6-1在FeT培养基上生长特征为:两株细菌均能形成0.05-0.1 mm的圆形菌落,菌落呈白色,透明状,其边缘齐整。两株细菌均为革兰氏阴性的好氧菌,菌体大小(长×宽):0.5-1.0μm×0.2-0.4 μm。两株细菌均能够利用苯丙氨酸和硫化氢,对尿素的利用呈弱阳性,但是对葡萄糖、乳糖、麦芽糖、甘露醇、甘露糖、蔗糖、阿拉伯糖、阿拉伯醇、D-核糖、木糖、木糖醇、半乳糖、松二糖、松三糖、山梨醇、山梨糖、蜜二糖、纤维二糖、葡萄糖铵、甲基-D葡萄糖苷、果糖、血清菊糖、DNA、酒石酸盐、葡萄糖酸盐、醋酸盐、卫茅醇(甜醇)、苦杏仁苷、鼠李糖、糊精、淀粉、肌醇、水杨素、β-半乳糖苷、丙二盐酸、西蒙氏枸橼酸盐、七叶苷、乙酰胺、硝酸盐(还原)、马尿酸盐、N-乙酰葡萄糖胺、丙酮酸钠等化合物不能够利用,不具有鸟氨酸脱羧酶、赖氨酸脱羧酶、精氨酸脱羧酶活性。在pH 2.0-11.0的范围两株细菌都能够生长,但是AMDY2-9-2最适生长pH值为5.0,AMDSY3-6-1最适生长pH值为6.0;两株细菌最适生长温度均为30℃,而且在NaCl含量为0%的FeT培养基中生长最好。AMDY2-9-2和AMDSY3-6-1与铁离子的作用机制研究表明,对于可溶性三价铁离子,两株细菌均需要完整的活细胞才能实现铁离子还原过程,单独作用的胞内活性酶、灭活的细胞和灭活的胞内酶均不能完成铁离子还原过程。两株细菌在不同三价铁离子浓度的FeT培养液中培养发现,与菌株AMDSY3-6-1比,菌株AMDY2-9-2生长比较快;对菌体与铁帽的相互作用研究结果表明,两株细菌均表现出还原速度慢的特点,详细的铁还原机制值得进一步研究。
[Abstract]:In this paper, two iron reducing bacteria isolated from the environment of Tongling acid mine in Anhui province were identified and studied. Two iron reducing microorganism (AMDY2-9-2 and AMDSY3-6-1) were isolated and purified from acidic mine environment in Tongling, Anhui Province by collecting the samples (acid ore water, tailings and draining ditch sediment), and using FeT culture medium to isolate and purify the two strains of iron reducing microorganism AMDY2-9-2 and AMDSY3-6-1. The 16SrDNA sequencing and homology analysis of the two microbes showed that the AMDY2-9-2 and A cidithiobacillus thiooxidans were the most close, the homology was 99.79, while AMDSY3-6-1 and Acidithiobacillus ferridurans were the most close, and the homology was 95.78. The growth characteristics of strain AMDY2-9-2 and AMDSY3-6-1 on FeT medium were as follows: both of them could form round colony of 0.05-0.1 mm, the colony was white and transparent, and its edge was even. The two strains were Gram-negative aerobic bacteria, and the cell size (length 脳 width) was 0.5-1.0 渭 m 脳 0.2-0.4 渭 m. Both strains were able to use phenylalanine and hydrogen sulfide and were weakly positive for urea use, but for glucose, lactose, maltose, mannitol, mannose, sucrose, arabinol D-ribose, xylitol, Galactose, pine disaccharide, trisaccharide, sorbitol, honey disaccharide, fibronectose, ammonium glucose, methyl D glucoside, fructose, serum chrysanthemum DNA, tartrate, gluconate, acetate, citronol (steviol), amygdalin, Rhamnose, dextrin, starch, inositol, salicylate, 尾 -galactoside, propanedioic acid, Simon's citrate, aescine, acetamide, nitrate (reduction), hippate N-acetyl glucosamine, sodium pyruvate, etc. No ornithine decarboxylase, lysine decarboxylase, arginine decarboxylase activity.
