斯氏油脂酵母细胞内油脂含量的快速检测方法研究

【摘要】:以斯氏油脂酵母为研究对象,探讨了利用苏丹黑B染色定量测定其胞内油脂含量的可行性,对苏丹黑B染色法测定胞内油脂含量的条件进行了优化。结果表明:斯氏油脂酵母经苏丹黑B染色处理后在600 nm处有最大吸收峰;在沸水浴中加热6 min和不添加盐酸的情况下,斯氏油脂酵母苏丹黑B染色效果最佳;建立了斯氏油脂酵母胞内油脂含量与吸光值A600的线性回归方程,线性关系显著(R2=0.992 9)。该方法与酸热-有机溶剂法测定结果相对误差在允许范围内,可用于快速测定斯氏油脂酵母胞内油脂含量。
[Abstract]:In order to STEVENSI oleaginous yeast as the research object, discusses the feasibility of using Sultan black B determination of fat content of its intracellular staining, intracellular determination of fat content of Sultan black B staining conditions were optimized. The results show that the STEVENSI oleaginous yeast by Sultan black B staining after treatment at 600 nm with maximum absorption peak; heating in a boiling water bath for 6 min and without adding hydrochloric acid under the condition of STEINERNEMATID oleaginous yeast Sultan black B staining effect is best established; oil content STEVENSI oil yeast cells and the light absorption value of A600 linear regression equation, a significant linear relationship (R2=0.992 9). The measurement results and the heat - acid organic solvent method, the relative error is within the allowable range, can be used for the rapid determination of oil content in STEVENSI oleaginous yeast intracellular.
