【摘要】:目的探讨使用甲壳素衍生物—羧化壳聚糖即刻分离制备血小板的可行性,寻找血液成分分离新方向。方法随机选取大连地区献血者40名,当日成功献血时同步采集全血10 m L,分成实验组,自然沉降对照组和离心对照组进行实验。从阿拉丁试剂公司购买水溶性壳聚糖,用血液保存液Ⅱ配制成(0.25~2.0)mg/m L不同浓度溶液,按1∶4比例加入到2 m L全血中,静置4 h,观察血液分离速度,计数血浆中红细胞,白细胞及血小板,观察血小板聚集现象及红细胞形态变化;在最适壳聚糖添加浓度的红细胞中加入适量红细胞保存液予以4℃保存,35 d保存期内检测溶血率。结果加入1.0~2.0 mg/m L浓度壳聚糖的实验组血液分离速度均明显快于自然沉降对照组,血浆中均仅存微量红细胞,富血小板,35d时无明显溶血,同离心对照组。结论壳聚糖可以用于采血后即刻分离富血小板血浆。
[Abstract]:Objective to investigate the feasibility of using chitin derivative carboxylated chitosan to prepare platelets and to find a new direction of blood component separation.
