miR-124抑制GluR2参与甲基苯丙胺PC12细胞成瘾

【摘要】:目的探讨mi R-124表达在甲基苯丙胺成瘾PC12细胞模型中的变化及其可能参与的调控机制。方法培养PC12细胞,随机分为6组:(1)正常对照组;(2)甲基苯丙胺组;(3)agomir Negative Control组;(4)mi R-124 agomir组;(5)agomir Negative Control+甲基苯丙胺组;(6)mi R-124 agomir+甲基苯丙胺组。处理结束后,提取细胞总RNA和蛋白,Real-time PCR检测miR-124表达,Western blot检测谷氨酸受体2亚单位(ionotropic glutamate receptor AMPA alpha 2,GluR 2)蛋白表达。结果与正常对照组相比,甲基苯丙胺处理组PC12细胞中miR-124表达明显降低,GluR 2蛋白表达明显升高;与agomir Negative Control+甲基苯丙胺组相比,miR-124agomir+甲基苯丙胺组PC12细胞中miR-124表达明显升高,GluR 2蛋白表达明显降低。结论 miR-124可能通过抑制GluR 2参与了甲基苯丙胺诱导的PC12细胞成瘾。
[Abstract]:Objective to investigate the changes of miR-124 expression in methamphetamine addicted PC12 cells and its possible regulatory mechanisms. Methods PC12 cells were cultured. They were randomly divided into 6 groups (1: 1) normal control group (2)) methamphetamine group (3) agomir Negative Control group, 4 agomir group, 5 agomir Negative Control methamphetamine group, 6 min R-124 agomir methamphetamine group, after the treatment, the control group was divided into 3 groups, the control group and the control group were randomly divided into 6 groups: 1) the control group (n = 6) was randomly divided into 6 groups (n = 6). The total RNA and real-time PCR of extracted cells were used to detect the expression of miR-124. Western blot was used to detect the expression of glutamate receptor-2 subunit (GluR2). Results compared with the normal control group, the expression of GluR2 protein was detected in glutamate glutamate receptor AMPA alpha 2G luR2. The expression of miR-124 decreased significantly in PC12 cells treated with methamphetamine. Compared with the agomir Negative Control methamphetamine group, the expression of miR-124 in PC12 cells was significantly increased and the expression of GluR2 protein was significantly decreased. Conclusion miR-124 may be involved in PC12 cell addiction induced by methamphetamine by inhibiting GluR 2.
