不同肿瘤细胞内源性DNA双链断裂水平与放射敏感性参数SF2的相关性

【摘要】:目的探讨不同肿瘤细胞内源性DNA双链断裂(DSBs)水平与放射敏感性参数SF2是否存在相关性。方法选取肺癌细胞系H1299、A549,乳腺癌细胞系MCF-7、MDA-MB-435s为研究对象。利用2 Gy射线照射后克隆形成实验检测不同细胞的放射敏感性指标SF2;通过实时荧光定量PCR方法测定不同细胞相对端粒长度变化;通过免疫荧光标记法标记不同肿瘤细胞γ-H2AX蛋白,检测不同细胞内源性DSBs损伤水平。通过统计学分析评价肿瘤细胞内源性DSBs与放射敏感性的关系。结果免疫荧光法标记γ-H2AX蛋白显示4种肿瘤细胞内源性DSBs水平肿瘤细胞除H1299与A549之间差异无统计学意义(P0.05)外,其他任意两组之间差异均有统计学意义(P0.05);4种肿瘤细胞的内源性DSBs水平与SF2、端粒长度的相关系数R分别为-0.95和-0.97,有统计学意义(P0.05)。结论肿瘤细胞内源性DSBs可以作为一种预测肿瘤细胞放射敏感性的标志,它们之间的相关性可能与端粒部分参与了内源性DSBs产生和修复有关。
[Abstract]:Objective to investigate the correlation between the levels of endogenous DNA double strand breaks (DSBs) and radiosensitivity parameters (SF2) in different tumor cells. Methods Lung cancer cell line H1299 A549 and breast cancer cell line MCF-7 MDA-MB-435s were selected. The radiosensitivity index SF _ 2 of different cells was detected by cloning assay after irradiation of 2 Gy, the relative telomere length of different cells was measured by real-time fluorescence quantitative PCR, and 纬 -H2AX protein was labeled by immunofluorescence labeling method. The levels of endogenous DSBs damage in different cells were detected. The relationship between endogenous DSBs and radiosensitivity of tumor cells was evaluated by statistical analysis. Results Immunofluorescence labeling of 纬 -H2AX protein showed that there was no significant difference between H1299 and A549 in the endogenous DSBs level of four kinds of tumor cells except H1299 and A549 (P 0.05). There were significant differences between other two groups in the level of endogenous DSBs and SF _ 2. The correlation coefficients of telomere length were -0.95 and -0.97, respectively. Conclusion Endogenous DSBs of tumor cells can be used as a marker to predict radiosensitivity of tumor cells. The correlation between them may be related to the part of telomere involved in the production and repair of endogenous DSBs.
