【摘要】:目的:通过检测维吾尔族饲鸽者肺外周血HLA-DQA1基因甲基化程度,并探讨其意义。方法:以20例确诊维吾尔族饲鸽者肺患者为病例组,20例维吾尔族饲养鸽子未发病者为阴性对照组,20例维吾尔族未饲养鸽子健康体检者为正常对照组。收集资料并抽取外周血标本,常规提取DNA、亚硫酸氢盐转化、PCR扩增、体外转录和RNase A特异性酶切,并行基质辅助激光解吸附电离飞行时间质谱分析技术检测HLA-DQA1基因甲基化情况。然后用spss17.0软件进行统计学分析,比较三组研究对象HLA-DQA1基因甲基化率是否存在差异,然后三组间两两比较HLA-DQA1甲基化率是否存在差异。结果:检测HLA-DQA1片段的Cp G位点总数4个(Cp G_1,Cp G_2,Cp G_3,Cp G_4),实际检测到的Cp G位点数3个(Cp G_2,Cp G_3,Cp G_4),Cp G_2、Cp G_4在三组样本中的甲基化率分布差异无统计学意义(p0.05)。Cp G_3在三组样本甲基化率分布差异具有统计学意义(p0.05)。维吾尔族饲鸽者肺患病组、维吾尔族饲养鸽子未发病组Cp G_3甲基化率高于维吾尔族未饲养鸽子健康体检组,差异具有统计学意义(P0.017)。但维吾尔族饲鸽者肺患病组与维吾尔族饲养鸽子未发病组Cp G_3甲基化率差异无统计学意义(P0.017)。结论:接触鸽子会影响HLA-DQA1基因甲基化率。但HLA-DQA1基因甲基化对维吾尔族饲鸽者肺的发病有无影响尚需进一步研究。
[Abstract]:Objective: to detect the degree of methylation of HLA-DQA1 gene in the peripheral blood of the Uygur pigeons and to explore its significance. Methods: 20 cases of Uygur pigeon lung patients were selected as the case group, 20 cases of Uygur people who were not infected with dove were negative control group, and 20 Uygur people who didn't keep dove health check-up as normal control group. Collect data and extract peripheral blood samples. Routine extraction of DNA, bisulfite transformation, PCR amplification, in vitro transcription and RNase A specific enzyme digestion, and matrix assisted laser desorption ionization time of flight mass spectrometry technology were used to detect the methylation status of HLA-DQA1 gene. Then SPSS17.0 software was used for statistical analysis. The difference of HLA-DQA1 gene methylation rate between the three groups was compared. Then the difference of HLA-DQA1 methylation rate between the three groups was compared between 22 groups. Results: the total number of Cp G loci detected in HLA-DQA1 fragments was 4 (Cp G_1, Cp G_2, Cp G_3, Cp G_4). The number of 3 G_3 spots was detected in the 3 groups. There was no significant difference in the methylation rate distribution between the two groups. The difference of the methylation rate of Cp G_3 in the three groups was statistically significant (P0.05). The Cp G_3 methylation rate of Uygur pigeon lung disease group and Uygur rearing dove group was higher than that of Uygur non dove health examination group, the difference was statistically significant (P0.017). However, there was no significant difference in the Cp G_3 methylation rate between the Uygur pigeon group and the Uygur dove uninfected group (P0.017). Conclusion: exposure to dove can affect the methylation rate of HLA-DQA1 gene. However, the effect of HLA-DQA1 gene methylation on the lung of the Uygur pigeon is still needed to be further studied.
