过继转移日本血吸虫感染鼠DC调控IL-13抑制过敏性哮喘黏液分泌的机制研究

【摘要】:目的探讨过继转移日本血吸虫感染鼠DC对IL-13细胞因子调控抑制过敏性哮喘黏液分泌的作用机制。方法采用MACS磁珠分选方法从日本血吸虫(SJ)感染小鼠及健康对照小鼠脾脏中提取DC,分别过继转移给BALB/c小鼠,并用OVA诱发哮喘。4周后处死小鼠,提取肺组织总RNA并制作病理切片,采取qRT-PCR和PAS染色方法等检测IL-13mRNA以及蛋白水平。建立DC与CD4+T细胞共培养体系,检测IL-13的表达情况。结果肺组织病理学检查显示,与OVA单纯哮喘组比较,过继转移SJ感染DC组(SJ+OVA)过敏性哮喘被抑制。qRT-PCR显示,过继转移SJ感染DC组IL-13mRNA表达受到抑制。PAS染色显示SJ感染DC组黏液分泌减少,免疫组化检查该组IL-13的表达量降低。DC与CD4+T细胞共培养结果显示SEA处理DC能抑制CD4+T细胞IL-13的表达。结论过继转移日本血吸虫感染鼠DC通过下调CD4+T细胞中IL-13细胞因子的释放而抑制过敏性哮喘的黏液分泌。
[Abstract]:Objective to investigate the mechanism of IL-13 cytokines regulating and inhibiting mucus secretion of allergic asthma in adoptive metastatic Schistosoma japonicum infected mice. Methods MACS magnetic beads were used to separate mice infected with Schistosoma japonicum and small healthy controls. DCS was extracted from mouse spleen and transferred to BALB/c mice. The mice were killed after 4 weeks of asthma induced by OVA. The total RNA of lung tissue was extracted and pathological sections were made. The levels of IL-13mRNA and protein were detected by qRT-PCR and PAS staining. The co-culture system of DC and CD4 T cells was established. Results Pulmonary histopathological examination showed that allergic asthma with adoptive SJ infection in DC group was inhibited by .qRT-PCR as compared with OVA asthma group. The inhibition of IL-13mRNA expression in DC infected with adoptive transfer of SJ showed that mucus secretion was decreased in DC infected with SJ. The expression of IL-13 was decreased by immunohistochemistry. The results of co-culture of DC and CD4 T cells showed that DC treated with SEA could inhibit the expression of IL-13 on CD4 T cells. Conclusion DC can down-regulate IL-13 fine in CD4 T cells of mice infected with schistosomiasis japonicum. The release of cytokines inhibits mucus secretion in allergic asthma.
