谷氨酰胺对克雷伯杆菌引起的大鼠肺炎炎症的抑制作用

【摘要】:目的探讨谷氨酰胺对克雷伯杆菌引起的大鼠肺炎炎症的抑制作用及机制。方法将36只SD大鼠随机分为3组,正常组、模型组、谷氨酰胺组,通过气管滴入克雷伯杆菌建立大鼠肺炎模型,谷氨酰胺组在造模前1d即腹腔注射10mL/kg谷氨酰胺,正常组和模型组给予等量生理盐水,连续给药7d,大鼠脱颈处死,取肺组织及血清进行检测。称量记录右肺湿重及干重(W/D),HE染色检测肺组织病理形态,酶联免疫吸附试验(ELISA)法检测肺组织匀浆及血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)含量,RT-PCR检测核因子-κB(NF-κB)p65和NF-κB抑制蛋白(IκBα)mRNA表达,Western blot检测NF-κB p6和IκBα蛋白表达。结果与模型组比较,谷氨酰胺组能降低W/D比值[5.98±0.29 vs.4.32±0.33(P0.05)],减轻肺组织充血水肿、炎性细胞浸润,改善肺组织形态,抑制血清和肺组织匀浆中TNF-α、IL-1β及IL-6的分泌(P0.05),并抑制NF-κB p65和IκBα磷酸化水平(P0.05)。结论谷氨酰胺能抑制克雷伯杆菌引起的大鼠肺炎炎症,与抑制NF-κB信号通路有关。
[Abstract]:Objective to investigate the inhibitory effect and mechanism of glutamine on pneumonia induced by Klebsiella in rats.Methods Thirty-six SD rats were randomly divided into 3 groups: normal group, model group and glutamine group. The model of pneumonia was established by instillation of Klebsiella in trachea. 10mL/kg glutamine was injected intraperitoneally in the glutamine group 1 day before the model was made.The normal group and the model group were given the same amount of normal saline for 7 days. The rats were killed without neck, and the lung tissue and serum were detected.The right lung wet weight and dry weight were measured by HE staining to detect lung histopathology.The expression of TNF- 伪 TNF- 伪, IL-1- 尾 -IL-1 尾 and IL-6IL-6 in lung homogenate and serum were detected by Elisa. The expression of NF-魏 B-NF- 魏 B)p65 and NF- 魏 B inhibitory protein I 魏 B 伪 mRNA was detected by RT-PCR. The expression of NF- 魏 B p6 and I 魏 B 伪 was detected by Western blot.Results compared with the model group, the glutamine group could reduce the W / D ratio [5.98 卤0.29 vs.4.32 卤0.33 vs.4.32 卤0.33 P0.05], alleviate the hyperemia and edema of lung tissue, inflammatory cell infiltration, and improve the morphology of lung tissue.It inhibited the secretion of TNF- 伪 IL-1 尾 and IL-6 in serum and lung tissue homogenate, and inhibited the phosphorylation levels of NF- 魏 B p65 and I 魏 B 伪.Conclusion Glutamine can inhibit the inflammation of pneumonia induced by Klebsiella in rats, which is related to the inhibition of NF- 魏 B signaling pathway.
