在树突状细胞中筛选与呼吸道合胞病毒诱发哮喘相关的长链非编码RNAs

【摘要】:目的使用人类全转录组芯片检测呼吸道合胞病毒(RSV)诱发哮喘中的树突状细胞(DCs),以筛选差异表达的长链非编码RNAs(lncRNAs)。在lncRNAs转录调控层面研究RSV诱发哮喘的机制。方法设立HDC-BEAS+RSV组、HDC-RSV组和HDC-CONTROL组3组,其中HDC-BEAS+RSV组是将RSV感染的人支气管上皮细胞BEAS-2B和未成熟DCs通过Transwell共培养,HDC-RSV组是将RSV感染未成熟DCs,HDCCONTROL组是将RSV对照液感染未成熟DCs。使用affymatrix的HTA2.0芯片检测3组DCs中基因和lncRNAs的表达差异,并对其进行聚类分析、GO分析和Pathway分析。结果筛选出有差异表达的25个基因和29个lncRNAs,其中20个基因表达上调,5个基因表达下调,27个lncRNAs表达上调,2个lncRNAs表达下调。上调的基因主要涉及促炎免疫反应,例如信号转导、趋化因子信号传导途径、细胞因子受体相互作用等。结论我们筛选出了RSV诱发哮喘中差异表达的lncRNAs,为进一步在lncRNA转录调控层面研究RSV诱发哮喘的机制奠定基础。
[Abstract]:Objective to detect dendritic cells (DC) in asthma induced by respiratory syncytial virus (RSVV) using human whole transcription microarray to screen differentially expressed long chain noncoding RNAsC RNAsS. To study the mechanism of asthma induced by RSV at the level of lncRNAs transcriptional regulation. Methods HDC-BEAS RSV group (HDC-RSV) and HDC-CONTROL group (3 groups) were established. The HDC-BEAS RSV group infects RSV infected human bronchial epithelial cells BEAS-2B and immature DCs through Transwell coculture. The HDC-BEAS RSV group infects immature DC-RSV with RSV and infects immature DC-RSV with RSV control fluid. The difference of gene and lncRNAs expression in three groups of DCs was detected by HTA2.0 chip of affymatrix, and then cluster analysis and Pathway analysis were carried out. Results Twenty-five differentially expressed genes and 29 lncRNAss were screened. Among them, 20 genes were up-regulated, 5 genes were down-regulated, 27 lncRNAs expressions were up-regulated and 2 lncRNAs expressions were down-regulated. The up-regulated genes are mainly involved in pro-inflammatory immune response, such as signal transduction, chemokine signaling pathway, cytokine receptor interaction and so on. Conclusion We screened out the differential expression of lncRNAsin in asthma induced by RSV and laid a foundation for further study on the mechanism of RSV induced asthma at the level of transcriptional regulation of lncRNA.
