BATF、RORγt及IL-17在哮喘小鼠脾组织中的表达及意义

【摘要】:目的探讨B细胞活化转录因子(BATF)、维甲酸相关核孤儿受体γt(RORγt)、白细胞介素-17(IL-17)在哮喘小鼠脾组织中的表达及意义。方法将15只小鼠随机分为3组:生理盐水(NS)组、哮喘(AS)组和地塞米松(DXM)组,每组5只。利用卵清蛋白(OVA)造模。收集支气管肺泡灌洗液(BALF)行白细胞分类计数;HE染色观察肺组织病理改变;酶联免疫吸附测定法(ELISA)检测脾组织匀浆上清液IL-17的质量浓度;实时荧光定量聚合酶链反应(RT-PCR)检测脾组织中BATF、RORγt mRNA的表达量;并对上述指标的相关性进行分析。结果 1)与AS组相比,DXM组小鼠BALF白细胞(WBC)总数、中性粒细胞(NEU)数和嗜酸性粒细胞(EOS)数明显降低(P0.01)。2)DXM组肺部炎症较AS组明显减轻。3)与AS组相比,DXM组脾组织匀浆上清液IL-17的质量浓度减低(P0.05)。4)与AS组相比,DXM组脾组织中BATF、RORγt mRNA表达量减少(P0.01)。5)脾组织匀浆上清液IL-17的质量浓度与BALF中WBC总数、NEU数、EOS数呈正相关(r=0.838、0.737、0.882,P均0.005);脾组织BATF mRNA的表达量与RORγt mRNA表达量、脾组织匀浆上清液IL-17的质量浓度呈正相关(r=0.807、0.833,P均0.005);脾组织RORγt mRNA的表达量与脾组织匀浆上清液IL-17的质量浓度呈正相关(r=0.890,P0.005)。结论 1)在哮喘小鼠脾组织中BATF与RORγt可能共同调节IL-17的分泌,并通过IL-17的致炎作用参与哮喘气道炎症的发生发展。2)DXM可能通过BATF、RORγt途径部分阻止脾源性Th17细胞的分化,减少IL-17的分泌。
[Abstract]:Objective to investigate the expression and significance of B cell activated transcription factor (BATFN), the orphan receptor 纬 t(ROR 纬 T of retinoic acid and interleukin-17 (IL-17) in the spleen of asthmatic mice. Methods Fifteen mice were randomly divided into three groups: NS group, ASA group and DXM group with 5 rats in each group. Ovalbumin (ovalbumin) was used to make the model. BALF was collected from bronchoalveolar lavage fluid (BALF) to observe the pathological changes of lung tissue by leucocyte classification and HE staining, and the IL-17 concentration in spleen tissue homogenate supernatant was detected by enzyme linked immunosorbent assay (Elisa). Real-time fluorescence quantitative polymerase chain reaction (RT-PCR) was used to detect the expression of BATFFU ROR 纬 t mRNA in splenic tissues, and the correlation of the above indexes was analyzed. Results 1) compared with as group, the total number of BALF WBC in DXM group was higher than that in as group. The number of neutrophils (neu) and eosinophilic granulocytes (EOS) were significantly reduced in the DXM group than in the as group. 3) the mass concentration of IL-17 in the splenic tissue homogenate supernatant of the DXM group was significantly lower than that in the as group. 4) the BATFROR 纬 t in the spleen tissue of the DXM group was significantly lower than that in the as group. The expression of mRNA in the supernatant of splenic tissue homogenate was positively correlated with the total number of WBC and the number of WBC in BALF (P 0.005), and the expression of BATF mRNA in splenic tissue was correlated with the expression of ROR 纬 t mRNA. There was a positive correlation between the mass concentration of IL-17 in the supernatant of spleen tissue and the expression of ROR 纬 t mRNA in spleen tissue and the mass concentration of IL-17 in splenic tissue homogenate supernatant. Conclusion 1) BATF and ROR 纬 t may co-regulate the secretion of IL-17 in the splenic tissues of asthmatic mice, and participate in the occurrence and development of asthma airway inflammation through the inflammatory effect of IL-17. 2 DXM may partially block the differentiation of splenic Th17 cells through the BATFU ROR 纬 t pathway. Reduce the secretion of IL-17.
