丹参对急性肺损伤血管生成素表达影响的实验研究

【摘要】:目的:探讨丹参在急性肺损伤中对血管生成素-1(angiopoietin-1,Ang-1)、血管生成素-2(angiopoietin-2,Ang-2)的调控作用及其对肺的保护机制。方法:36只清洁级SD雄性大鼠随机分为生理盐水对照组(NS组)、LPS组和丹参干预组。(1)对照组12只,每只大鼠从尾静脉注入相同剂量的生理盐水。(2)LPS组12只,,每只从尾静脉注入LPS(5mg/kg)造成急性肺损伤模型组。(3)丹参干预组12只,每只尾静脉注入LPS(5mg/kg)建模,1小时后再从尾静脉注入丹参注射液10g/kg(正大青春宝药业公司生产)。每组实验大鼠在12小时后处死,收集肺泡灌洗液、血液、肺组织进行肺湿/干重(W/D)比值、肺系数(LI)、肺通透系数(LPI)等的计算;HE染色观察大鼠肺组织病理改变;免疫印记法测Ang-1、Ang-2在肺组织的表达。结果:LPS组及丹参组肺组织的湿/干重(W/D)比值、肺系数(LI)、肺通透系数(LPI)均较对照组明显升高,差异具有统计学意义(p0.01),且病理切片都有明显的炎症损伤改变。Ang-1在对照组蛋白表达量较LPS组及丹参组明显增高(p0.05),Ang-2蛋白表达量在LPS组和丹参组较对照组蛋白表达量明显增多(p0.01)。LPS组与丹参组相比,湿/干重(W/D)比值、肺系数(LI)、肺通透系数(LPI)较丹参组升高明显(p0.05)。结论:在急性肺损伤中,肺组织中Ang-2与Ang-1的表达调控维持肺毛细血管的稳定起着重要作用。丹参能使急性肺损伤肺组织中Ang-2的表达减少,Ang-1的表达增多来使毛细血管的渗出减少减轻炎症所致的肺间质水肿、红细胞漏出来达到肺保护作用。
[Abstract]:Aim: to investigate the effects of salvia miltiorrhiza on angiopoietin-1 Ang-1 and angiopoietin-2 Ang-2 in acute lung injury. Methods Thirty-six clean SD male rats were randomly divided into control group (n = 12) and salvia miltiorrhiza intervention group (n = 12). Acute lung injury model group was induced by injecting 5 mg / kg LPS5 mg / kg into tail vein. 12 rats were treated with salvia miltiorrhiza (LPS 5 mg / kg) and then 10 g / kg of salvia miltiorrhiza injection was injected into caudal vein for 1 hour after injection of LPS5 mg / kg into caudal vein of Salvia miltiorrhiza (Zhengyangbao Pharmaceutical Company). The rats in each group were killed 12 hours later. The lung wet / dry weight (W / D) ratio, lung coefficient (Li), lung permeability coefficient (LPI), lung permeability index (LPI), lung tissue were collected from alveolar lavage fluid, blood and lung tissue to observe the pathological changes of lung tissue. The expression of Ang-1 Ang-2 in lung tissue was measured by immunoblotting. Results compared with the control group, the wet / dry weight W / D ratio, lung coefficient Li, lung permeability index of lung tissue in the control group were significantly higher than those in the control group. The expression of Ang-1 in the control group was significantly higher than that in the LPS group and the salvia miltiorrhiza group. Compared with Salvia miltiorrhiza group, Compared with Salvia miltiorrhiza group, the wet / dry weight W / D ratio, lung coefficient, lung permeability coefficient and lung permeability coefficient were significantly higher than those of Danshen group (P < 0.05). Conclusion: the expression of Ang-2 and Ang-1 plays an important role in maintaining the stability of pulmonary capillaries in acute lung injury. Salvia miltiorrhiza can reduce the expression of Ang-2 in lung tissue of acute lung injury and increase the expression of Ang-1 so that the capillary exudation can reduce the interstitial edema caused by inflammation and the red blood cells leak out to achieve lung protection.
