老年慢性阻塞性肺疾病中硫氧还蛋白系统及谷胱甘肽系统表达的变化及意义

【摘要】:目的:研究硫氧还蛋白系统中的关键功能蛋白硫氧还蛋白1(TRX1)、硫氧还蛋白样2(TRXL-2)、硫氧还蛋白还原酶(TRXR)和谷胱甘肽系统中的还原型谷胱甘肽1(GSH1)、γ-谷胱甘肽半胱氨酸合成酶(γ-GCS)在老年慢性阻塞性肺疾病(COPD)中的表达变化,初步探讨硫氧还蛋白系统及谷胱甘肽系统在老年COPD中的抗氧化作用。 方法:收集不同年龄组的GOLD分级C级稳定期的老年COPD病人和对照组的肺组织,及外周血和痰液。采用免疫组化实验检测GSH1、TRX1、TRXL-2、TRXR、γ-GCS在肺组织中的表达。收集老年COPD病人及对照组的外周血及晨痰,采用Western blot实验检测血细胞中GSH1、TRX1、TRXL-2、TRXR、γ-GCS的表达水平,采用ELISA实验检测血浆、血清及痰液中GSH1、TRX1、TRXL-2、TRXR、γ-GCS的含量。 结果:免疫组化实验结果显示GSH1、TRX1、TRXL-2、TRXR、γ-GCS在肺组织细胞中均有表达,Western blot实验结果显示血细胞中同样存在目标蛋白的表达,ELISA实验结果显示目标蛋白在血浆、血清及痰液中也有表达。Western blot和ELISA实验结果表明:在血细胞、血浆、血清及痰液上清中,与对照组相比,老年COPD组GSH1、TRX1、TRXL-2、TRXR、γ-GCS的表达水平或含量均显著上调(P 0.05)。COPD组内,随着年龄的增加,目标蛋白的表达水平或含量均显著下降(P 0.05)。实验结果提示在老年COPD疾病中,,机体硫氧还蛋白系统与谷胱甘肽系统的抗氧化应激功能增强,但随着年龄的增加,两大系统的抗氧化应激功能逐渐降低。 结论: 硫氧还蛋白系统及谷胱甘肽系统参与了老年慢性阻塞性肺疾病的发病机制,为从硫氧还蛋白系统及谷胱甘肽系统角度研究老年性COPD疾病诊断、治疗手段提供了新的思路
[Abstract]:Objective: to study the key functional proteins in thioredoxin system, thioredoxin 1 (TRX1), thioredoxin (2T), thioredoxin reductase (TRXR) and glutathione 1 (GSH 1), 纬-glutathione cysteine synthase (GST) in the thioredoxin system. Expression of 纬 -GCS in elderly patients with chronic obstructive pulmonary disease (COPD), To investigate the antioxidant effects of thioredoxin system and glutathione system in elderly COPD. Methods: lung tissue, peripheral blood and sputum were collected from elderly patients with GOLD grade C stable stage and control group. The expression of TRXL 2 TRXR, 纬 -GCS in lung tissue was detected by immunohistochemistry. Peripheral blood and morning sputum were collected from elderly patients with COPD and control group. The expression levels of TRXL 2 TRXR, 纬 -GCS in blood cells were detected by Western blot test. The contents of TRXL 2 TRXR, 纬 -GCS in plasma, serum and sputum were detected by ELISA test. Results: the results of immunocytochemistry showed that GSH1TX1 TRXL-2 TRXR, 纬 -GCS were all expressed in lung tissue cells. Western blot assay showed that there was the same expression of target protein in blood cells. The results of Elisa showed that the target protein was in plasma. The results of Western blot and ELISA in serum and sputum showed that the expression level or content of GSH1TX 1 TRXL 2 TRXL 2 TRXR, 纬 -GCS in blood cell, plasma, serum and sputum supernatant were significantly up-regulated in P 0.05).COPD group with the increase of age, compared with the control group, the expression level or content of GSH1TX 1 TRXL 2 TRXL 2 TRXR, 纬 -GCS were significantly up-regulated in P 0.05).COPD group. The expression level or content of target protein decreased significantly (P 0.05). The results suggested that the antioxidant stress function of thioredoxin system and glutathione system was enhanced in the aged patients with COPD disease, but the antioxidant stress function of the two systems decreased gradually with the increase of age. Conclusion: Thioredoxin system and glutathione system are involved in the pathogenesis of chronic obstructive pulmonary disease in the elderly, which provides a new idea for the diagnosis and treatment of senile COPD disease from the perspective of thioredoxin system and glutathione system.
