芹菜素逆转白介素6诱导的中性粒细胞凋亡延迟

【摘要】:目的:观察芹菜素对炎性因子白细胞介素6(interleakin-6,IL-6)刺激的中性粒细胞(neutrophils,Neu)凋亡的影响,并探讨可能的分子机制。方法:采取健康志愿者外周静脉血,分离纯化Neu,随机分为对照组、芹菜素组、IL-6组、IL-6+芹菜素组及加入广谱天冬氨酸特异性半胱氨酸蛋白酶(caspase)抑制剂的Q-VD-Oph+IL-6+芹菜素组、z-VAD-fmk+IL-6+芹菜素组。流式细胞术观察Neu凋亡率,Western blot检测Neu活化的caspase-3(cleaved caspase-3)的表达情况。结果:与对照组比较,芹菜素促进Neu凋亡(P0.05),IL-6抑制Neu凋亡(P0.05);两者共孵育时,芹菜素可以部分逆转IL-6诱导的Neu凋亡延迟(P0.05);与IL-6组比较,芹菜素与IL-6共孵育组cleaved caspase-3的蛋白表达水平显著增高(P0.05),而且芹菜素对caspase-3的活化作用和对IL-6诱导的Neu凋亡延迟的逆转作用可以被caspase抑制剂消除(P0.05)。结论:芹菜素可能通过caspase途径逆转IL-6诱导的Neu凋亡延迟,为今后治疗Neu炎症性气道疾病提供了新思路。
[Abstract]:Aim: to investigate the effect of apigenin on neutrophil (neutrophils Neu) apoptosis induced by interleukin 6 (IL 6) interleakin-6 (IL 6) and to explore the possible molecular mechanism. Methods: Neu was isolated and purified from peripheral venous blood of healthy volunteers. It was randomly divided into control group, IL-6 group and Q-VD-Oph IL-6 apigenin group added with broad-spectrum aspartate specific cysteine protease caspase inhibitor, z-VAD-fmk IL-6 apigenin group. The apoptosis rate of Neu and the expression of caspase-3(cleaved caspase-3 activated by Neu were detected by flow cytometry and Western blot. Results: compared with the control group, apigenin promoted the apoptosis of Neu and inhibited the apoptosis of Neu by P0.05IL-6. When incubated with apigenin, the apoptosis of Neu induced by IL-6 was partially reversed by apigenin, and compared with that of IL-6 group. The expression level of cleaved caspase-3 protein was significantly increased in cleaved caspase-3 co-incubated with apigenin and IL-6. Moreover, the activation of caspase-3 by apigenin and the reversal of Neu apoptosis induced by IL-6 could be eliminated by caspase inhibitor. Conclusion: apigenin may reverse the delayed apoptosis of Neu induced by IL-6 via caspase pathway and provide a new idea for the treatment of Neu inflammatory airway disease.
