α-半乳糖神经酰胺对小鼠肺iNKT细胞数量、活性和气道炎症反应的影响

【摘要】:目的观察α-半乳糖神经酰胺(α-GalCer)对小鼠肺iNKT细胞数量、活性和气道炎症反应的影响。方法 12只野生型Balb/c小鼠随机分为α-GalCer治疗组和正常对照组,每组6只;12只CD1d~(-/-)Balb/c小鼠随机分为CD1d~(-/-)α-GalCer治疗组和CD1d~(-/-)对照组,每组6只。野生型和CD1d~(-/-)α-GalCer治疗组小鼠单次腹腔注射α-GalCer 2μg,并用等量PBS对照。分别采用流式细胞仪检测小鼠肺iNKT细胞、IL-4~+和IFN-γ~+i NKT细胞的数量;HE和PAS染色观察肺组织炎症和气道杯状细胞增生;姬姆萨染色检测肺泡灌洗液(BALF)细胞总数和分类计数;ELISA法检测BALF中IL-4、IL-5、IL-10、IL-13水平和脾细胞培养上清液中IL-4、IL-5、IL-13水平。结果野生型α-GalCer治疗组小鼠肺iNKT细胞、IL-4~+和IFN-γ~+i NKT细胞的数量明显高于野生型正常对照组(P0.05)。野生型α-GalCer治疗组小鼠肺组织无炎症改变和基底膜杯状细胞增生;BALF中细胞总数和分类计数、BALF及脾细胞培养上清液中IL-4、IL-5和IL-13水平与野生型对照组比较差异无统计学意义(P0.05)。野生型α-GalCer治疗组小鼠BALF中IL-10水平明显高于野生型正常对照组(P0.01),但CD1d~(-/-)α-GalCer治疗组小鼠BALF中IL-10水平与野生型正常对照组和CD1d~(-/-)对照组比较差异无统计学意义(均P0.05)。结论腹腔注射α-GalCer可以增加小鼠肺iNKT细胞数量和活性,但不能诱导气道炎症反应。
[Abstract]:Objective to investigate the effects of 伪 -galactose-ceramide (伪 -GalCer) on the number, activity and airway inflammation of iNKT cells in mice. Methods Twelve wild-type Balb / c mice were randomly divided into 伪 -GalCer treatment group and normal control group. Six mice in each group were randomly divided into two groups: CD1d- / (-r-) 伪 -GalCer treatment group and CD1d- (-r-) control group with 6 mice in each group. Mice in wild-type and CD1d- 伪 -GalCer group were given a single intraperitoneal injection of 伪 -GalCer 2 渭 g, and the same dose of PBS was used as control. The number of IL-4 ~ and IFN- 纬 ~ I NKT cells were detected by flow cytometry. The inflammation and goblet cell proliferation of the airway were observed by HE and pas staining. The total number of alveolar lavage fluid (BALF) cells and their classification count were detected by Giemsa staining. The levels of IL-4, IL-5, IL-10 and IL-13 in BALF and spleen cell culture supernatant were detected by Elisa. Results the number of IL-4 ~ and IFN- 纬 ~ I NKT cells in the wild-type 伪 -GalCer treated group was significantly higher than that in the wild-type normal control group (P0.05). There were no inflammatory changes in lung tissue and the total number and classification of BALF cells in the wild-type 伪 -GalCer treated mice. There was no significant difference in the levels of IL-4 IL-5 and IL-13 in the supernatant of spleen cell culture between the wild-type 伪 -GalCer group and the wild-type control group (P0.05). The level of IL-10 in BALF of wild-type 伪 -GalCer treated mice was significantly higher than that of wild-type normal control group (P0.01), but the level of IL-10 in BALF of wild-type 伪 -GalCer treated group was not significantly different from that of wild-type normal control group and CD1d- / (-) control group (P0.05). Conclusion Intraperitoneal injection of 伪 -GalCer can increase the number and activity of iNKT cells, but can not induce airway inflammation.
