烟曲霉菌提取物对人支气管上皮细胞修复功能的影响

【摘要】:目的探讨烟曲霉菌对呼吸道上皮细胞修复功能的影响及其可能机制。方法实验分为正常对照组、不同浓度(10、20、40 mg/L)AFE组、热处理AFE组。分别应用不同浓度和热处理的烟曲霉菌提取物(aspergillus fumigatus extract,AFE)作用体外培养的人支气管上皮细胞16HBE不同的时间,以细胞体外损伤修复功能、细胞迁移能力、定量细胞黏附能力、细胞伸展面积为指标,观察AFE和热处理的AFE对上皮细胞修复能力的影响。结果20、40 mg/L AFE均明显抑制16HBE细胞的体外损伤修复功能、细胞迁移能力和细胞黏附能力(P0.05),并呈明显的时间和浓度依赖性。10 mg/L AFE与正常对照组差异无统计学意义(P0.05)。10、20、40 mg/L AFE均明显抑制细胞的伸展能力(P0.05),并呈明显浓度依赖性。另外,热处理AFE的蛋白酶完全灭活,对上述观察指标无明显影响(P0.05)。结论 AFE依赖其蛋白酶活性明显抑制细胞的修复功能,并呈明显的浓度和时间依赖性。
[Abstract]:Objective to investigate the effect of Aspergillus fumigatus on the repair function of respiratory epithelial cells and its possible mechanism. Methods the experiment was divided into normal control group, AFE group with different concentration (10 ~ 20 mg / L) and heat treated AFE group. Different concentrations and heat treatment of Aspergillus fumigatus extract (aspergillus fumigatus extract AFE) were used to treat 16HBE of human bronchial epithelial cells cultured in vitro for different time. The cell damage repair function, cell migration ability and cell adhesion ability were measured in vitro. The effects of AFE and heat treated AFE on the repair ability of epithelial cells were observed. Results 40 mg / L AFE significantly inhibited the repair function of 16HBE cells in vitro. The cell migration ability and cell adhesion ability (P0.05) were significantly time-and concentration-dependent. 10 mg / L AFE had no significant difference compared with the normal control group (P0.05). 10 mg / L AFE significantly inhibited the cell extension (P0.05), and in a dose-dependent manner. In addition, the protease of heat treatment AFE was completely inactivated, which had no significant effect on the above indexes (P0.05). Conclusion AFE dependent protease activity significantly inhibited the repair function of the cells in a concentration and time dependent manner.
