谷氨酰胺对急性肺损伤大鼠组织修复的影响

【摘要】:目的肺是多器官功能障碍中最容易受累的器官,严重感染、创伤、休克、酸中毒及有害气体吸入等多种因素均可诱发急性肺损伤(acute lung injury, ALI),其是临床上常见的导致危及生命的急性呼吸衰竭原因之一。本研究通过对脓毒症诱导急性肺损伤大鼠模型研究,探讨谷氨酰胺对脓毒症大鼠急性肺损伤时的肝细胞生长因子(HGF)及角质细胞生长因子(KGF)的作用,以期进一步探讨谷氨酰胺对肺损伤后组织修复的影响。 方法采用改良盲肠结扎穿刺法复制大鼠脓毒症急性肺损伤模型,40只健康SD大鼠,分为3组,即对照组、手术组[改良盲肠结扎穿刺法]、谷氨酰胺治疗组[采用改良盲肠结扎穿刺法后静脉注射3%谷氨酰胺0.75g/kg]。实验中手术组大鼠死亡6只,治疗组大鼠死亡3只,在手术6h后手术组随机处死6只,24h后处死剩下5只,治疗组药物处理6h后处死6只,24h后处死剩下的8只,留取标本行血气分析,肺干湿重测量,4000r/min离心10min,收集血清,置于-20℃冰箱保存待用。Elisa法测定血清中HGF和KGF的含量。并进行各组肺组织的病理学光镜检查。研究谷氨酰胺对急性肺损伤大鼠组织修复的影响。 结果 1.HGF和KGF的比较:与对照组相比较,手术组及治疗组在术后6小时、24小时HGF及KGF含量明显升高(p0.05),治疗组术后6小时、24小时HGF及KGF含量较手术组明显升高(p0.05),手术组术后6小时与24小时相比较,HGF、KGF含量在6h、24h间差异无显著性(P0.05)。 2.血气结果的比较:与对照组比较,动脉氧分压(partial pressureof oxygen in artery, PaO2)在改良盲肠结扎穿刺术后6小时、24小时各组均有下降(P0.05);在术后6小时、24小时,手术组较同时间点谷氨酰胺治疗组下降的差异有统计学意义(P0.05); 3.肺干湿重的比较:实验组、治疗组在术后6小时、24小时的肺湿干重比值与对照组之间存在增高表现(p0.05),手术组较治疗组在术后6小时、24小时肺湿干重比均明显增高(P0.05)。 结论 1.静脉给予谷氨酰胺能促进急性肺损伤大鼠血清中HGF、KGF浓度水平的上升,从而促进受损组织的修复。
[Abstract]:Objective the lung is one of the most easily involved organs in multiple organ dysfunction, with severe infection, trauma and shock. Acidosis and inhalation of harmful gases can induce acute lung injury (acute lung injury, Ali), which is one of the common causes of life-threatening acute respiratory failure. In this study, we investigated the effects of glutamine on hepatocyte growth factor (HGF) and keratinocyte growth factor (KGF) during acute lung injury induced by sepsis in rats. To further explore the effect of glutamine on tissue repair after lung injury. Methods Forty SD rats with septic acute lung injury model were established by modified cecal ligation puncture method and divided into 3 groups: control group. Operation group [modified cecal ligation puncture method], glutamine treatment group [modified cecum ligation puncture method after intravenous injection of 3% glutamine 0.75g/kg]. In the experiment, 6 rats died in the operation group, 3 rats died in the treatment group, 5 rats were killed at random in the operation group after 6 hours of operation, 6 rats in the treatment group were killed after 6 hours of drug treatment, and 8 rats were killed after 24 hours of drug treatment. The samples were collected for blood gas analysis, the lung dry-wet weight was measured by 4000r / min centrifugation for 10 mins, and the serum was collected and stored in -20 鈩,
