乌司他丁对脂多糖诱导大鼠急性肺损伤中NF-κBp65及MMP-2表达的影响
[Abstract]:AIM: To investigate the expression of nuclear factor-kappa B (NF-KB) p65 and matrix metalloproteinase-2 (MMP-2) in lung tissues of rats with acute lung injury (ALI) induced by lipopolysaccharide (LPS), and to explore the signal transduction of NF-kappa B. Meanwhile, by observing the lung pathology, the changes of NF-kappa B p65 and MMP-2 in ALI rats after ulinastatin intervention, the protective effect of ulinastatin on LPS-induced ALI and its possible mechanism were investigated.
Methods: Forty-five healthy male SD rats were randomly divided into normal control group, acute lung injury group and ulinastatin intervention group, 15 rats in each group. (1) Normal control group: tail vein injection of normal saline (2ml/kg). (2) ALI group: tail vein injection of lipopolysaccharide (5mg/kg) induced ALI rat model. (3) Ulinastatin intervention group: rats were intraperitoneally injected with ulinastatin. (100ku / kg), 30 minutes later, the tail vein was injected with lipopolysaccharide (5mg / kg). The rats were sacrificed 4 hours after the tail vein administration and the corresponding pulmonary tissue and blood samples were taken. The arterial oxygen partial pressure (PaO2) was measured and the pulmonary tissue was taken for examination. Wet-to-dry weight ratio (W/D) was measured. The lung tissue was observed with naked eyes. The pathological changes of HE staining were observed under light microscope. The expressions of NF-KBp65 and MMP-2 in lung tissue were detected by immunohistochemistry.
Results: 1. Changes of PaO2: Pa02 fluctuated around 103.1 mmHg in the normal control group; Pa02 in the ALI group was significantly lower than that in the normal control group (P 0.05); Pa02 in the Ulinastatin intervention group was higher than that in the ALI group, but still lower than that in the normal control group (P 0.05). W/D fluctuated around 4.33; W/D in lung tissue of ALI group increased significantly compared with normal control group (P 0.05); W/D in lung tissue of ulinastatin intervention group was lower than that of ALI group, but still higher than that of normal control group. 3. Compared with normal control group, the expression of NF-kappa Bp65 and MMP-2 in lung tissue of ALI group and Ulinastatin intervention group increased significantly (P 0.05). The expression of NF- kappa Bp65 and MMP-2 in the intervention group was lower than that in the ALI group (P0.05).
Conclusion: 1. The expression of NF-kappa B p65 and MMP-2 in the lung tissue of LPS-induced acute lung injury rats increased; 2. Ulinastatin can inhibit the expression of NF-kappa B p65 and MMP-2 in the lung tissue of acute lung injury rats; 3. Ulinastatin may have protective effect on LPS-induced acute lung injury rats.
