抗IL-1β和TNF-α IgY抗体治疗豚鼠过敏性支气管哮喘的机制

【摘要】:目的:探讨抗IL-1β和TNF-αIgY抗体雾化吸入治疗豚鼠过敏性支气管哮喘的机制。方法:Hartly豚鼠随机分为雾化吸入生理盐水的正常对照组(C组),以及雾化吸入卵清白蛋白(OVA)建立过敏性哮喘模型的哮喘模型组(M组)、1.0%抗IL-1β和TNF-αIgY模型豚鼠治疗组(Z1组)和布地奈德混悬液模型豚鼠治疗组(Z2组)。各组豚鼠末次治疗结束后2、4、8、24 h心脏穿刺取血;收集支气管肺泡灌洗液(BALF)。美蓝及伊红染色计数血液嗜酸性粒细胞、嗜中性粒细胞、淋巴细胞、巨噬细胞、嗜碱性粒细胞。ELISA法测定血浆和BALF中IL-1β、IL-4、IL-6、IL-8、IL-13、IL-16、TNF-α、TGF-β1、IgE的含量。结果:Z1组嗜酸性粒细胞在2~8 h低于M组。Z1组血浆IL-1β(4、24 h)、IL-16(2、4、24 h)、TGF-β1(4~24 h)、IgE(24 h)较M组显著降低(P0.05),而在BALF中IL-1β和TNF-α(2~8 h),IL-4、IL-6、IL-8和IL-13(4~24 h),IL-16(8 h)、TGF-β1和IgE(4~8 h)显著降低于M组,尤其是IL-1β和TNF-α在2 h就开始显著降低(P0.05)。IL-1β及TNF-α与IL-4、IL-6、IL-8、IL-13、IL-16、TGF-β1、IgE等呈正相关(P0.05)。结论:抗IL-1β和TNF-αIgY雾化吸入减轻OVA诱导的豚鼠过敏性哮喘炎症可能是通过降低多种过敏性炎症因子浓度而实现。
[Abstract]:Objective: to investigate the mechanism of anti-IL-1 尾 and anti-TNF- 伪 IgY aerosol inhalation in the treatment of allergic bronchial asthma in guinea pigs. Methods Guinea pigs were randomly divided into normal control group (group C) and asthmatic model group (group M) induced by atomized ovalbumin (OVA) (group M), which were treated with 1.0% anti IL-1 尾 and TNF- 伪 IgY (group Z1). Guinea pig model treated with budesonide suspension (group Z 2). After the last treatment in each group, the blood samples were taken from the heart for 24 hours after 2 minutes and 8 minutes. The bronchoalveolar lavage fluid (BALF).) was collected from the guinea pigs with bronchoalveolar lavage fluid. Blood eosinophils, neutrophils, lymphocytes, macrophages and basophil were counted by methylene blue and eosinophil staining. Elisa method was used to detect the content of IL-1 尾 -IL-4, IL-6, IL-8, IL-13, TNF- 伪, TGF- 尾 _ 1 and IgE in plasma and BALF. Results the levels of IL-1 尾 (424 h) and TGF- 尾 1 (424 h) in group Z1 were significantly lower than those in group M (P 0.05), while the levels of IL-1 尾 and TNF- 伪 (2h) in BALF were significantly lower than those in group M (P 0.05). The levels of IL-1 尾 and TNF- 伪 (2h) in BALF and IL-13 (424 h) TGF- 尾 1 and IgE (4h) were significantly lower than those in group M (P 0.05). In particular, IL-1 尾 and TNF- 伪 began to decrease significantly at 2 h (P0.05). IL-1 尾 and TNF- 伪 were positively correlated with IL-4, IL-6, IL-8, IL-13, IL-16, TGF- 尾 1-IgE and so on (P0.05). Conclusion: the anti IL-1 尾 and TNF- 伪 IgY aerosol inhalation may reduce the inflammation of allergic asthma induced by OVA in guinea pigs by reducing the concentration of many kinds of allergic inflammatory factors.
