矽肺患者肺泡巨噬细胞表面A类清道夫受体表达及其对细胞凋亡的调控作用

【摘要】:目的探讨矽肺患者肺泡巨噬细胞(AM)表面A类清道夫受体(SR-A)的表达及其对AM凋亡的调控作用,为矽肺的防治提供依据。方法以不同期别的矽肺患者51例为调查对象,以4例矽肺观察对象为对照组,收集其大容量肺灌洗回收液,分离、纯化AM,将纯化后的AM分为5组:未处理组、酪氨酸激酶抑制剂组、活性氧抑制剂组、抗-MARCO组和抗-MSR1组,37℃处理24h后,裂解各组AM,采用Western blot方法检测受体蛋白MSR1和MARCO及凋亡蛋白Caspase-3的表达并计算其相对水平。结果矽肺患者AM中Caspase-3的表达水平均高于对照组,且随着肺纤维化程度的加重逐渐增高(P0.05)。MSR1和MARCO表达水平虽有随矽肺期别增高而增加的趋势,但差异无统计学意义(P0.05)。未脱尘组Caspase-3的表达水平高于脱尘组(P0.05);MSR1、MARCO表达水平与Caspase-3均呈正相关。抗-MARCO和抗-MSR1抗体处理后,矽肺患者Caspase-3的表达均高于未处理组,贰期矽肺患者MSR1表达高于未处理组,贰期和叁期矽肺患者MARCO的表达均较未处理组上调(P0.05)。贰期、叁期矽肺患者的AM经N-乙酰半胱胺酸处理后Caspase-3表达水平下调。结论 SR-A在介导矽肺患者AM凋亡中可能发挥重要的作用。
[Abstract]:Objective to investigate the expression of class A scavenger receptor (SR-A) on alveolar macrophages (AM) and its role in regulating AM apoptosis in silicosis patients, and to provide evidence for the prevention and treatment of silicosis.
