Pre-miRNA单核苷酸多态性与慢性阻塞性肺疾病的相关性研究

【摘要】:研究目的: 采用病例-对照研究方法探索pre-miRNA基因多态性与中国甘肃地区汉族人群慢性阻塞性肺疾病之间的相关性。包括pre-miRNA基因中SNP位点rs2910164G/C、rs11614913C/T、 rs3746444C/T。 研究方法: 在兰州大学第一医院和四川大学华西医院筛选532例没有血缘关系的临床诊断明确的COPD患者和511例正常对照人群。收集COPD患者及正常对照人群血液样本,采用离心柱法提取样本的全基因组DNA,用紫外分光光度仪测定DNA的浓度,然后设计位点特异性扩增引物,利用聚合酶链式反应和限制性片段长度多态性(PCR-RFLP)的方法分析了3个位于pre-miRNA基因的单核苷酸位点(rs11614913, rs3746444, and rs2910164)在中国汉族人群COPD患者和正常人中的等位基因和基因型频率,使用统计学软件SPSS11.5分析两者的差异。 结果: SNP位点rs11614913C/T和rs3746444A/G的等位基因频率和基因型频率在病例组和对照组之间的分布有显著的差异。rs11614913C/T多态性的CC基因型和C等位基因和COPD发病增高显著相关(CC对比IT, OR=0.55,95%CI,0.372-0.818;C对比T,OR=0.806,95%CI,0.676-0.962),类似的,rs3746444A/G多态性的AA基因型和A等位基因也表现出了与COPD发病增高很强的相关性(AA对比GG,OR=0.158,95%CI,0.046-0.542;A对比G, OR=0.733,95%CI,0.569-0.943).然而,本研究的另外一个多态性位点:rs2910164G/C的等位基因频率和基因型频率在病例组和对照之间的并没有表现出明显的差异。 结论: 研究显示rs11614913C/T和rs3746444A/G多态性在COPD的发病过程中起到重要作用,提示pre-miRNA hsa-mir-196a和hsa-mir-499基因在中国汉族人群中与COPD的发病有很强的相关性,而pre-miRNA hsa-mir-146a并没有表现出与COPD发病的相关性。
[Abstract]:Objective: to investigate the association between pre-miRNA gene polymorphism and chronic obstructive pulmonary disease (COPD) in Han population in Gansu province of China. Including SNP rs2910164G/C,rs11614913C/T, rs3746444C/T. in the pre-miRNA gene Methods: 532 COPD patients and 511 normal controls were selected from the first Hospital of Lanzhou University and Huaxi Hospital of Sichuan University. Blood samples from patients with COPD and normal controls were collected. The concentration of DNA was determined by UV spectrophotometry, and then locus specific primers were designed. Polymerase chain reaction (PCR) and restriction fragment length polymorphism (PCR-RFLP) were used to analyze three single nucleotide loci (rs11614913, rs3746444,) of pre-miRNA gene. And rs2910164) alleles and genotype frequencies in COPD patients and normal controls were analyzed by SPSS11.5 software. Results: the allele frequency and genotype frequency of rs11614913C/T and rs3746444A/G at SNP locus were significantly different between case group and control group. CC genotype and C allele and COPD were increased in rs11614913C/T polymorphism. High significant correlation (CC contrast IT,) OR=0.55,95%CI,0.372-0.818; Similarly, the AA genotypes and A alleles of rs3746444A/G polymorphisms also showed a strong correlation with the increased incidence of COPD (AA versus GG,OR=0.158,95%CI,). 0.046-0.542; A contrast G, OR=0.733,95%CI,0.569-0.943). However, there was no significant difference in allelic and genotypic frequencies between the case group and the control group at another polymorphic locus in this study: rs2910164G/C. Conclusion: rs11614913C/T and rs3746444A/G polymorphisms play an important role in the pathogenesis of COPD, suggesting that pre-miRNA hsa-mir-196a and hsa-mir-499 genes are strongly associated with COPD in Chinese Han population. Pre-miRNA hsa-mir-146a did not show a correlation with the onset of COPD.
