TNF-α、VEGF和MMP-9在大鼠自身免疫性肺气肿发病中的作用

【摘要】:目的:探讨肿瘤坏死因子-α(TNF-a)、血管内皮生长因子(VEGF)、基质金属蛋白酶-9(MMP-9)在大鼠自身免疫性肺气肿中的作用,为研究其发病机制及寻求新的治疗方法提供理论依据。 方法:清洁级SD大鼠20只随机分为2组:佐剂对照组(n=10)和自身免疫性肺气肿模型组(简称模型组,n=10)。将原代培养的人脐静脉内皮细胞及佐剂注入大鼠腹腔内建立自身免疫性肺气肿模型,佐剂对照组只给予佐剂大鼠腹腔内注射。两组大鼠均取肺组织切片行苏木素-伊红染色(HE)观察病理学改变,并定量测定肺平均内衬间隔(MLI)、平均肺泡数(MAN),以ELISA检测支气管肺泡灌洗液(BALF)中的TNF-a、VEGF和MMP-9含量。 结果:(1)模型组MLI比佐剂对照组增高,而其MAN比佐剂对照组降低,差异均有统计学意义(P均0.05)。 (2)模型组BALF中MMP-9含量较佐剂对照组升高(P0.05),而两组TNF-α含量差异无统计学意义(P0.05)。 (3)模型组BALF中VEGF含量较佐剂对照组降低(P0.05),差异有统计学意义。 结论:(1)将脐静脉内皮细胞注入正常大鼠腹腔即可以建立自身免疫性肺气肿模型。 (2)MMP-9含量升高、VEGF含量减低可能参与大鼠自身免疫性肺气肿的形成,而TNF-a与该病的形成无关。
[Abstract]:Objective: to investigate the role of tumor necrosis factor-伪 (TNF-a) and vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9) in autoimmune emphysema in rats. It provides a theoretical basis for the study of its pathogenesis and the search for new therapeutic methods. Methods: twenty clean grade SD rats were randomly divided into two groups: adjuvant control group (n = 10) and autoimmune emphysema model group (n = 10). The model of autoimmune emphysema was established by injecting primary cultured human umbilical vein endothelial cells and adjuvant into the abdominal cavity of rats. The adjuvant control group was given only intraperitoneal injection of adjuvant rats. The pathological changes of lung tissue were observed by hematoxylin-eosin staining (HE), and the mean alveolar number (MAN),) of mean lining interval (MLI),) was measured quantitatively. The TNF-a, in bronchoalveolar lavage fluid (BALF) was detected by ELISA. VEGF and MMP-9 contents. Results: (1) the MLI of the model group was higher than that of the adjuvant control group, while the MAN of the model group was lower than that of the adjuvant control group. (2) the content of MMP-9 in BALF of the model group was higher than that of the adjuvant control group (P0.05), but there was no significant difference between the two groups in the content of TNF- 伪 (P0.05). (3) the content of VEGF in BALF of model group was lower than that of adjuvant control group (P0.05). Conclusion: (1) the model of autoimmune emphysema can be established by injecting umbilical vein endothelial cells into the abdominal cavity of normal rats. (2) the increase of MMP-9 content and the decrease of VEGF content may be involved in the formation of autoimmune emphysema in rats, but TNF-a is not related to the formation of autoimmune emphysema.
