实验性肺纤维化小鼠循环单核细胞亚群的动态变化及意义

【摘要】:目的外周血单核细胞表型偏移失衡与肺纤维化病理进展关系密切。文中通过阐述循环单核细胞亚群在实验性肺纤维化小鼠不同病理时期的动态变化,探讨其表型偏移与肺组织炎症和纤维化的关系。方法 100只雄性C57BL/6J小鼠随机数字表法分为等渗盐水组和博莱霉素组,经口咽吸入法分别给予等渗盐水及博莱霉素A5,术后第1、3、7、14及21天处死小鼠,病理学方法计算各组小鼠肺组织炎症评分(inflammation score,IS)及胶原容积分数(collagen volume fraction,CVF),常规方法进行支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)细胞计数及分类计数,荧光定量PCR法检测各组小鼠肺组织胶原Ⅰ、胶原Ⅲ的mRNA表达量,氯胺T法检测肺组织羟脯氨酸(hydroxyproline,HYP)含量,流式细胞术检测循环单核细胞亚群的变化。结果博莱霉素组小鼠第3天炎性细胞浸润、肺泡壁水肿等炎症程度(IS评价)较等渗盐水组显著增加(P0.01),第7天达到峰值后呈下降趋势。博莱霉素组小鼠第14、21天肺组织CVF较等渗盐水组明显增高(P0.01);与等渗盐水组相应时间点比较,博莱霉素组第1天细胞总数无明显变化,第3天明显增多(25.18±2.06 vs 8.82±1.59,P0.01),第7天升至高峰(56.56±5.03 vs 3.68±0.79,P0.01),后呈下降趋势,但第14~21天细胞总数仍高于等渗盐水组(P0.01),肺泡巨噬细胞比例的变化趋势与细胞总数相一致;而中性粒细胞数量第1、3、7天显著高于等渗盐水组(9.086±1.268 vs 1.108±0.229、5.551±0.511 vs 0.315±0.100、8.093±0.922 vs 0.249±0.074),差异均有统计学意义(P0.01);博莱霉素组胶原Ⅰ、ⅢmRNA表达水平在第14、21天显著高于等渗盐水组(P0.05);HYP检测结果显示博莱霉素组第7、14、21天高于等渗盐水组(P0.01),其中第21天达最高值;与等渗盐水组相比,博莱霉素组第1天Ly6Chi单核细胞亚群比例显著升高,第3天达到高峰(P0.01),此后呈降低趋势,Ly6Clo亚群比例变化与之相反。各组小鼠Ly6Chi单核细胞比例与IS及CVF值均呈正相关(P0.01,P=0.001 3)。结论在博莱霉素A5诱导的小鼠肺纤维化模型中,不同病理生理阶段外周血中Ly6Chi、Ly6Clo单核细胞亚群比例处于动态变化,与肺组织炎性细胞渗出、浸润等病理进程相比,Ly6Chi单核细胞亚群应对肺组织损伤、炎症反应迅速,早期Ly6Chi单核细胞亚群比例的升高可能与肺组织炎症反应及后期纤维化程度存在密切的联系。
[Abstract]:Objective the imbalance of phenotypic deviation of peripheral blood monocytes is closely related to the pathological progress of pulmonary fibrosis. In this paper, the dynamic changes of circulating monocyte subsets in different pathological stages of experimental pulmonary fibrosis mice were described, and the relationship between phenotypic deviation and inflammation and fibrosis of lung tissue was discussed. Methods 100 male C57BL/6J mice were randomly divided into isoosmotic saline group and bleomycin A5 group. The mice were sacrificed on the 1st, 3rd, 7th, 14th and 21st day after operation.
