烟草烟雾暴露对人中性粒细胞诱捕网形成的影响及红霉素干预作用的体外研究

目的:探讨不同浓度香烟烟雾提取物浓度在体外对外周血中性粒细胞生成中性粒细胞外诱捕网(NETs)的影响,吸烟相关慢性阻塞性肺疾病患者较正常人对烟草烟雾刺激外周血中性粒细胞产生NETs敏感性的异同及红霉素的干预作用。方法:招募平均年龄在64岁的稳定期COPD患者及健康自愿者各10名,分离纯化其外周血中性粒细胞后进行刺激分组:(1)分别使用0.1%、0.3%、0.5%、1%及2%浓度的香烟烟雾提取物(CSE)对正常人中性粒细胞进行刺激,利用荧光DNA定量法观察各浓度CSE刺激下中性粒细胞发生NETosis的情况;(2)使用PMA、CSE分别对COPD组及正常人组其进行刺激,应用免疫荧光染色法观察各组NETs的生成情况及CSE诱导生成NETs的形态特征及组成结构;应用荧光DNA定量法对各组中性粒细胞的NETs生成情况进行定量;(3)对正常人外周血中性粒细胞在固定使用0.1%CSE进行刺激的前提下,分别用浓度为1μg/mL、5μg/mL及10μg/mL的红霉素对其进行干预,利用荧光DNA定量法定量各浓度红霉素干预下对烟草烟雾诱导中性粒细胞发生NETosis化的抑制情况;(4)在固定使用0.1%C...
