IL7R基因多态性与汉族人群哮喘相关性研究

研究背景:哮喘是由于遗传和环境因素所导致的一种复杂的慢性气道疾病,遗传度为30-70%。随着全基因组关联分析方法大规模的应用,越来越多的哮喘易感基因被鉴定,但由于人种的差异,这些结果需要在不同的人群中进行重复和验证。2006年,一个德国研究小组利用定位候选克隆方法在5p13区域发现了包括IL7R在内的多个哮喘易感位点。由于该区域在多个人群中已反复被证实是一个与哮喘连锁的一个区域,而且在这个区域内含有多个细胞因子相关的基因。鉴于以上证据,加之考虑到IL7R的功能,我们认为IL7R是一个哮喘的候选基因。 目的:本研究的主要目的是利用基于群体的病例对照关联分析方法探讨白细胞介素7受体基因在汉族人群哮喘发生中的作用。 材料和方法:收集哮喘患者及正常对照血样并提取基因组DNA,所收集的400个哮喘患者均符合支气管哮喘诊断标准,424个正常对照均无过敏性哮喘及其他过敏性疾病的症状及病史并排除肝、肾疾病。全部标本来自潍坊哮喘病医院及山东大学齐鲁医院,病例组个体与对照组个体均为年龄和性别匹配的山东汉族人。 利用Haploview分析来自HapMap数据库中HCB人群的IL7R基因的数据,我们从中选择了...
