HoxA13在各型尿道下裂尿道板及包皮中的分布及意义

【摘要】：目的：研究各种类型的尿道下裂其尿道板和包皮中的HoxA13基因转录及其蛋白表达,探讨在尿道下裂形成过程中HoxA13的作用。方法：按前、中、后三型分型尿道下裂,每型各10例患儿,取其包皮及尿道板,年龄3.3岁-11.6岁。取10例包茎患儿包皮作为对照,年龄3.1岁～10.4岁。取10例阴茎癌尿道作为尿道板对照,年龄48.1岁～75.6岁。免疫组织化学(IHC)和逆转录PCR (RT-PCR)检测相应组织中HoxA13的蛋白表达量和转录水平。结果：RT-PCR:HoxA13转录水平在对照组(尿道：1.270±0.209,包皮：1.409±0.441)高于中间型(尿道板：0.684±0.228,包皮：0.848±0.338)和后型尿道下裂(尿道板：0.464±0.164,包皮：0.497±0.218)(P0.05)；前型(尿道板：1.054±0.189,包皮：1.071±0.342)转录水平高于后型(P0.05)。免疫组织化学：对照组(尿道13420±2636,包皮：12050±4112)HoxA13蛋白表达水平高于中间型(尿道：5238±3065,包皮：5217±1993)和后型(尿道板：2238±2217,包皮：2095±1591)(P0.05)；前型(尿道板：10450±2123,包皮：8223±3212)表达水平高于后型(P0.05)。结论：HoxA13在尿道下裂患者包皮和尿道板组织中的表达程度可能与尿道下裂严重程度有关。
[Abstract]:Aim: to study the transcription and protein expression of HoxA13 gene in urethral plate and prepuce of hypospadias and to explore the role of HoxA13 in the formation of hypospadias. Methods: according to the classification of hypospadias according to the type of anterior, middle and posterior, 10 children with hypospadias were divided into three types. The prepuce and urethral plate of 10 children with prepuce and urethra, aged from 3.3 to 11.6 years, were taken as control group. The urethra of 10 cases of penile carcinoma, aged 48.1 years and 75.6 years, were used as control. The expression of HoxA13 protein and transcription level in the corresponding tissues were detected by immunohistochemistry and reverse transcription PCR RT-PCR. Results in the control group (urethral: 1.270 卤0.209, prepuce: 1.409 卤0.441), the transcription level of: RT-PCR: HoxA13 in the control group was higher than that in the intermediate type (urethral plate: 0.684 卤0.228, prepuce: 0.848 卤0.338) and posterior hypospadias (urethral plate: 0.464 卤0.164, prepuce: 0.497 卤0.218), and the transcription level in the former type (urethral plate: 1.054 卤0.189, prepuce: 1.071 卤0.342) was higher than that in the posterior type (P0.05). Chemistry: the expression levels of HoxA13 in the control group (urethral 13420 卤2636, prepuce 12050 卤411212) were higher than those in the intermediate type (urethra: 5238 卤3065, prepuce: 5217 卤1993) and the posterior type (urethral plate: 2238 卤2217, prepuce: 2095 卤1591p0. 05), and in the former type (urethral plate 10 450 卤2123, prepuce: 8 223 卤3212) were higher than those in the posterior type (urethral plate: 2238 卤2217, prepuce: 10450 卤2123, prepuce: 8223 卤3212). Conclusion the expression of w HoxA13 in the prepuce and urethral plate of hypospadias may be related to the severity of hypospadias.
