PSCA基因多态性与膀胱癌易感性关系的Meta分析

【摘要】:目的:进一步探讨前列腺干细胞抗原(PSCA)基因rs2294008多态性与膀胱癌易感性的关系。方法:系统检索PubMed、MEDLINE、EMBASE、Cochrane图书馆、中国生物医学数据库(CBM)、相关期刊论文(CNKI)及万方数据库中2016年10月之前的有关PSCA基因rs2294008多态性与膀胱癌易感性相关的研究。严格制定纳入及排除标准,提取数据,采用NOS量表进行质量评价,应用Stata12.0软件进行Meta分析,应用OR值及其95%CI评估其关联强度,并进行发表偏倚评估分析。结果:共纳入文献8篇,包括13个病例-对照研究,涉及12349病例及24836例对照。Meta分析结果显示:在5种基因模型中PSCA rs2294008(CT)多态性与膀胱癌易感性均显著相关,进一步按种族及病例来源进行亚组分析,等位基因频率模型(T vs C)[OR=1.151,95%CI=1.12-1.19,P=0.0000.001,I~2=0.0%];显性模型(CT+TT vs CC)=[OR=1.271,95%CI=1.20-1.33,P=0.0000.001,I~2=0.0%]、隐性模型(TT vs CC+CT)=[OR=1.151,95%CI=1.08-1.21,P=0.0000.001,I~2=0.0%]、纯合子模型(TT vs CC)=[OR=1.311,95%CI=1.22-1.40,P=0.0000.001,I~2=0.0%]及共显性模型(CT vs CC)=[OR=1.25,95%CI=1.18-1.32,P=0.0000.001,I~2=0.0%],以上模型分析结果均说明:PSCA rs2294008(CT)多态性与膀胱癌显著相关。结论:PSCA rs2294008(CT)基因多态性与膀胱癌的易感性显著相关,且与种族及病例来源无关;突变型T等位基因可能为膀胱癌的易感基因位点,PSCA有望成为膀胱癌遗传易感性的生物学标记物。
[Abstract]:Objective: to investigate the relationship between rs2294008 polymorphism of prostate stem cell antigen (PSCA) gene and bladder cancer susceptibility.Methods: a systematic search was conducted on the PSCA gene rs2294008 polymorphism and its association with bladder cancer susceptibility in PubMedus MEDLINE Cochrane Library, Chinese Biomedical Database (CBM), Chinese Journal Full-text Database (CNKI) and Wanfang database before October 2016.The inclusion and exclusion criteria were strictly formulated, the data were extracted, the quality was evaluated by NOS scale, the Meta was analyzed by Stata12.0 software, the correlation strength was evaluated by OR value and its 95%CI, and the evaluation of publication bias was carried out.Results: a total of 8 articles were included, including 13 case-control studies, involving 12349 cases and 24836 controls.
