磷酸甘油酸变位酶1在生精功能障碍睾丸组织中的表达及意义

【摘要】:目的:检测磷酸甘油酸变位酶1(PGAM1)在不同程度生精功能不良睾丸组织中的表达情况,并分析PGAM1表达与临床病理参数的关系。方法:收集生精功能正常、轻度生精功能不良、重度生精功能不良、唯支持细胞综合征患者睾丸组织标本,统计性激素六项结果,采用免疫组化、qRT-PCR检测PGAM1在不同程度生精功能不良睾丸组织中的表达情况。结果:免疫组化结果显示PGAM1的高表达率在生精功能正常、轻度生精功能不良、重度生精功能不良及唯支持细胞综合征患者各组间比较,差异具有统计学意义(P=0.000),PGAM1表达与患者的年龄、睾丸体积、LH及T表达均无明显相关(P0.05);而PGAM1低表达患者FSH水平高于PGAM1高表达患者(P=0.002 3);qRT-PCR结果提示PGAM1 mRNA在生精功能正常及唯支持细胞综合征患者睾丸组织中表达水平明显高于重度生精不良睾丸组织中的表达,差异具有统计学意义(P=0.000),PGAM1 mRNA表达与患者的年龄、睾丸体积、LH及T表达均无明显相关(P0.05),而在FSH≤12 mIU/mL患者睾丸组织中的表达明显高于FSH12 mIU/mL患者(P=0.043 0)。结论:PGAM1的表达与睾丸生精功能障碍有关,其低表达与重度生精功能不良的形成相关,其异常高表达与唯支持细胞综合征的形成有关。
[Abstract]:Aim: to detect the expression of phosphoglyceric acid mutase 1 (PGAM1) in testis with different degrees of spermatogenic dysfunction and to analyze the relationship between the expression of PGAM1 and clinicopathologic parameters. Methods: normal spermatogenic function, mild spermatogenic dysfunction and severe spermatogenic dysfunction were collected from testicular tissues of patients with Sertoli cell syndrome. Immunohistochemical qRT-PCR was used to detect the expression of PGAM1 in testicular tissues with different degrees of spermatogenic dysfunction. Results: Immunohistochemical results showed that the high expression rate of PGAM1 was compared among the patients with normal spermatogenic function, mild spermatogenic dysfunction, severe spermatogenic dysfunction and Sertoli cell syndrome. The difference was statistically significant between the expression of PGAM1 and the age of the patients. There was no significant correlation between the expression of LH and T in testicular volume, while the FSH level in patients with low expression of PGAM1 was higher than that in patients with high expression of PGAM1. The results of RT-PCR showed that the expression of PGAM1 mRNA in testicular tissues of patients with normal spermatogenic function and Sertoli's syndrome was clear.
