选择性雄激素受体调节剂(Sarm)对迟发性腺功能减退大鼠的作用研究

【摘要】:目的通过观察选择性雄激素受体调节剂对迟发性腺功能减退症(LOH)大鼠的作用,为治疗LOH提供理论依据。 方法将30只符合标准的老年雄性Wistar大鼠作为LOH动物模型,并随机分成3组:对照组(A)、DHT组(B)、Sarm组(C),分别给予等计量的空白溶剂、雄激素及选择性雄激素受体调节剂(Sarm)进行干预,实验4周后观察各组大鼠LOH症状的改善情况并评分,记录大鼠悬尾及强迫游泳时间,检测血清游离睾酮的水平,计算前列腺指数。 结果干预4周后A、B、C三组大鼠LOH症状评分分别为(18.20±1.87)、(30.00±2.49)、(35.20±1.87),C组优于B组,B组优于A组,差异有统计学意义;悬尾不动时间(s)分别为(196.17±10.57)、(167.29±2.15)、(160.67±6.92),C组短于B组,B组短于A组,差异有统计学意义;强迫游泳时间(min)分别为(16.33±1.16)、(24.50±1.26)、(30.43±0.71),C组长于B组,B组长于A组,差异有统计学意义;血清游离睾酮(pg/mL)分别为(2.248±0.305)、(1.088±0.258)、(2.526±0.283),B组低于A组和C组,差异有统计学意义;前列腺指数(%)分别为(0.321±0.008)、(0.419±0.015)、(0.172±0.007),B组高于A组而C组低于A组,各组间差异有统计学意义(P0.01)。 结论Sarm可通过其与雄激素受体高度亲和力及选择性调节雄激素受体表达,明显改善LOH模型大鼠的LOH症状,有效抑制前列腺组织的增生,不影响自身睾酮的分泌。
[Abstract]:Objective to observe the effect of selective androgen receptor modulator on delayed hypogonadism in rats and to provide a theoretical basis for the treatment of LOH. Methods Thirty old male Wistar rats were randomly divided into three groups: control group (control group) and control group. The control group was treated with equal dose of blank solvent, androgen and selective androgen receptor modulator. After 4 weeks of experiment, the improvement of LOH symptoms and scores were observed, the time of tail suspension and forced swimming were recorded, the level of serum free testosterone was measured, and the prostate index was calculated. Results after 4 weeks of intervention, the scores of LOH symptoms in group C were 18.20 卤1.87, 30.00 卤2.49 and 35.20 卤1.87, respectively, which were significantly better than those in group B, and the time of suspension immobility was 196.17 卤10.577.29 卤2.150.67 卤6.92, respectively, which was significantly shorter than that in group B (P < 0.05). The time of forced swimming was 16.33 卤1.16, respectively (24.50 卤1.26). Group C was longer than group B (30.43 卤0.71) than group B, the difference was statistically significant, the serum free testosterone pg / mL was 2.248 卤0.305 and 1.088 卤0.2588 卤0.2583, respectively, and the difference was statistically significant. The prostatic index was 0.321 卤0.008 (0.419 卤0.015) in group B was higher than that in group A (0.172 卤0.007), but lower in group C than in group A (P 0.01). Conclusion Sarm can regulate androgen receptor expression through its high affinity and selectivity with androgen receptor, improve LOH symptoms of LOH model rats, inhibit prostatic hyperplasia effectively, and do not affect the secretion of self testosterone.
